摘 要:以维多利亚蓝B(victoria blue B,VBB)为探针分子,利用表面增强拉曼散射(surface enhanced Raman scattering,SERS)结合酶催化反应构建氧乐果快速检测方法。结果表明,在水浴温度为37℃,将乙酰胆碱酯酶活化15 min,再进行酶催化反应15 min,反应后静置10 min,滴加金纳米溶胶后再静置15 min,在检测体系中乙酰胆碱酯酶、碘化乙酰硫代胆碱、金纳米溶胶、VBB的浓度分别为5μg/mL、5μmol/L、0.195μg/mL、0.25μmol/L条件下,建立的检测体系在1614 cm-1处的SERS吸收峰值△I与3.5 ng/L~403.5 ng/L的氧乐果浓度呈良好的线性关系,相关系数为R2=0.9958,检出限为1.90×10-4 ng/L。当常见离子和农药在体系中共存时,对氧乐果的测定不产生干扰,说明体系稳定性良好。加标回收率为99.02%,相对标准偏差为1.17%。A rapid detection method for omethoate was established based on surface-enhanced Raman scattering(SERS)and enzyme-catalyzed reaction with victoria blue B(VBB)as the probe.The results showed that after the activation of acetylcholinesterase for 15 min and reaction for 15 min in the water bath at 37℃,standing for 10 min,dropping of gold nanoparticles(AuNPs),and standing for 15 min,the concentrations of acetylcholin esterase(AchE),acetylthiocholine iodide(ATI),AuNPs,and VBB were 5μg/mL,5μmol/L,0.195μg/mL,and 0.25μmol/L,respectively.The SERS absorption peak△I of the established reaction system at 1614 cm-1 showed a good linear relationship with the concentration of omethoate within the range of 3.5 ng/L-403.5 ng/L,with R2=0.9958 and the detection limit of 1.90×10-4 ng/L.The coexistence of common ions and pesticides in the system did not interfere with the detection of omethoate,which indicated that the system had good stability.The results of standard addition experiment showed the recovery rate of 99.02%and the relative standard deviation of 1.17%.
