摘 要:目的:探究miR-495-3p调控BUB1表达水平对胆管癌细胞增殖和侵袭的影响及机制。方法:qRT-PCR法测定人肝内正常胆管上皮细胞(HIBEpiC)、胆管癌细胞RBE、IHC-ST1、SNU-869、HUCCT1中miR-495-3p和BUB1mRNA表达水平;将SNU-869细胞分为空白对照组、阴性对照组、miR-495-3pmimics组、miR-495-3p mimics+pcDNA组、miR-495-3p+pc-BUB1组,比较各组SNU-869细胞增殖、迁移和侵袭能力情况、miR-495-3p及BUB1 mRNA表达水平以及BUB1、ki-67、Bcl-2、MMP-2蛋白表达情况。结果:与HIBEpiC细胞相比,RBE、IHC-ST1、SNU-869、HUCCT1细胞中miR-495-3p表达水平降低(P<0.05),BUB1表达水平升高(P<0.05)。与空白对照组相比,miR-495-3p mimics组SNU-869细胞中miR-495-3p表达升高(P<0.05),增殖能力、侵袭细胞数、迁移细胞数、ki-67、MMP-2、BUB1表达降低(P<0.05),Bcl-2蛋白表达水平升高(P<0.05);与miR-495-3p mimics组比较,miR-495-3p+pc-BUB1组SNU-869细胞中BUB1表达升高(P<0.05),增殖能力、侵袭细胞数、迁移细胞数、ki-67、MMP-2表达升高(P<0.05),Bcl-2蛋白表达水平降低(P<0.05)。结论:过表达miR-495-3p可显著抑制SNU-869细胞增殖、迁移和侵袭能力,可能通过下调BUB1蛋白表达发挥作用。Objective: To explore the effects and mechanism of miR-495-3p regulating the expression of BUB1 on the proliferation and invasion of cholangiocarcinoma cells. Methods: qRT-PCR method was used to determine the expression levels of miR-495-3p and BUB1 mRNA in human normal intrahepatic bile duct epithelial cells HIBEpiC, cholangiocarcinoma cells RBE, IHC-ST1, SNU-869, and HUCCT1;the SNU-869 cells were divided into blank control group, negative control group,miR-495-3p mimics group, miR-495-3p mimics+pcDNA group, and miR-495-3p+pc-BUB1 group,the proliferation, migration and invasion ability, the expression levels of miR-495-3p, BUB1 mRNA,and the protein expression BUB1, ki-67, Bcl-2, and MMP-2 in SNU-869 cells among each groups were compared. Results: Compared with that respectively in HIBEpiC cells, the expression level of miR-495-3p in RBE, IHC-ST1, SNU-869, and HUCCT1 cells was reduced(P<0. 05), the expression level of BUB1 was increased(P<0. 05). Compared with that respectively in the blank control group, the expression of miR-495-3p in SNU-869 cells in the miR-495-3p mimics group was increased(P<0. 05), the proliferation ability, the number of invasive cells, the number of migrating cells, the expression of ki-67, MMP-2, and BUB1 were decreased(P<0. 05), the expression level of Bcl-2protein was increased significantly(P<0. 05);compared with that respectively in the miR-495-3p mimics group, the expression of BUB1 in SNU-869 cells of the miR-495-3p+pc-BUB1 group was increased(P<0. 05), the proliferation ability, the number of invasive cells, the number of migrating cells, the expression of ki-67, MMP-2, and BUB1 were increased(P<0. 05), and the expression level of Bcl-2 protein was reduced significantly(P<0. 05). Conclusion: Overexpression of miR-495-3p can significantly inhibit the proliferation, migration, and invasion of SNU-869 cells, which may play a role by down-regulating the expression of BUB1 protein.
