摘  要:为明确苦荞醋饮中的主要化学成分,并进一步探讨其作用机制,该研究采用超高效液相色谱-四极杆飞行时间串联质谱(UPLC-Q-TOF-MS/MS)技术分析苦荞醋饮中的化学成分,运用HIT 2.0、Drugbank 5.0、DAVID等数据库对苦荞醋饮发挥治疗“三高”的靶点及通路进行分析,并通过Cytoscape软件构建网络。结果表明,采用UPLC-Q-TOF-MS/MS技术共鉴定出109个化学成分,其发挥降压、降脂、降糖作用的“成分-靶点-疾病”网络包含34个活性成分、286个药物靶点。京都基因和基因组百科全书(KEGG)富集分析获得179条通路。“关键成分-关键靶点-信号通路-疾病”网络分析结果显示,苦荞醋饮可能以木犀草素、槲皮素等为关键成分,作用于细胞肿瘤抗原p53(TP53)、RAC-α丝氨酸/苏氨酸蛋白激酶(AKT1)等关键靶点,主要参与调控磷脂酰肌醇3激酶-蛋白激酶B(PI3K-Akt)等信号通路,发挥降压、降脂、降糖作用。To clarify the main chemical components in tartary buckwheat vinegar beverage,and to further explore its action mechanism,the chemical components of tartary buckwheat vinegar beverage were analyzed by ultra-performance liquid chromatography-quadrupole-time of flight mass spectrometry(UPLC-Q-TOF-MS/MS).The targets and pathways of tartary buckwheat vinegar in"hypertension,hyperlipemia and hyperglycemia"treatment were analyzed via HIT 2.0,Drugbank 5.0,DAVID and other databases,and the network was constructed by Cytoscape software.The results showed that a total of 109 chemical components were identified by UPLC-Q-TOF-MS/MS technology,the"component-target-disease"network which had the exerting antihypertensive,lipid-lowering and glucose-lowering effects contained 34 active components and 286 drug targets.A total of 179 pathways were identified from Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis.The network analysis results of"key component-key target-signaling pathway-disease"showed that components in tartary buckwheat vinegar such as luteolin and quercetin,might be the key components which acted on the key targets such as cellular tumor antigen p53(TP53)and RAC-alpha serine/threonine-protein kinase(AKT1).These components mainly involved in the regulation of phosphoinositide 3 kinases-protein kinase B(PI3K-Akt)and other signaling pathways,and exerted antihypertensive,lipid-lowering and glucose-lowering effects.
