摘 要:为了筛选出用于犬细菌性皮肤病的新型替抗产品,本研究在采集的泡菜样本中分离乳酸菌菌株,通过琼脂扩散法检测乳酸菌菌株是否可以分泌抗菌活性物质,并通过初筛与复筛确定试验菌株。利用形态学观察与16S rRNA测序方法对受试乳酸菌菌株进行鉴定。通过酸碱稳定试验、热稳定试验及水解酶稳定试验,研究该菌株所产细菌素的生物学特性;并以犬细菌性皮肤病的主要病原菌金黄色葡萄球菌、伪中间型葡萄球菌、绿脓杆菌、大肠杆菌为指示菌进行细菌素抑菌谱的测定。结果表明:从泡菜样本中分离得到的3株乳酸菌菌株,经过初筛及复筛最终获得1株乳酸菌满足后续试验,将其命名为LP-3。通过形态学观察及16S rRNA基因序列测定,LP-3为平房乳杆菌。LP-3所产细菌素具有良好的热稳定性,在酸性环境中抗菌活性较强,可以被蛋白酶K水解。其所产细菌素对金黄色葡萄球菌、伪中间型葡萄球菌、绿脓杆菌、大肠杆菌均有良好的抑菌效果。综上,该平房乳杆菌细菌素对犬细菌性皮肤病主要病原菌具有良好的抑菌作用,为研究防治犬细菌性皮肤病的新型药物奠定基础。In order to screen out new alternative antibiotics for bacterial skin diseases in dogs,in this study,lactic acid bacteria strains were isolated from kimchi samples collected,and AGAR diffusion method was used to detect whether lactic acid bacteria strains could secrete antibacterial active substances,and the test strains were determined by preliminary screening and re-screening.The strains were identified by morphological observation and 16S rRNA sequencing.The biological characteristics of bacterin produced by the strain were studied by acid-base stability test,heat stability test and hydrolase stability test.The antibacterial spectrum of bacteriocin was determined with Staphylococcus aureus,Staphylococcus pseudointermedia,Pseudomonas aeruginosa and Escherichia coli as the main pathogens of bacterial skin diseases in dogs.The results showed that three lactic acid bacteria strains were isolated from kimchi samples,and one strain of lactic acid bacteria was finally obtained after preliminary screening and re-screening to meet the follow-up test,which was named LP-3.Through morphological observation and 16S rRNA gene sequencing,LP-3 was identified as Lactobacillus hirsutum.The bacteriocin produced by LP-3 has good thermal stability and strong antibacterial activity in acidic environment,and can be hydrolyzed by protease K.The bacteriocin has a good antibacterial effect on Staphylococcus aureus,Staphylococcus Pseudointermediate,Pseudomonas aeruginosa and Escherichia coli.In conclusion,the bacteriocin of lactobacillus has a good bacteriostatic effect on the main pathogenic bacteria of canine bacterial skin disease,which lays a foundation for the research of new drugs for prevention and treatment of canine bacterial skin disease.
