摘 要:在金针菇全基因组中鉴定到4个蓝光受体基因和1个红光受体基因。分别对5个基因的序列进行生物信息学分析,包括基因一级结构分析、保守结构分析、亚细胞定位预测以及跨膜区分析。亚细胞定位结果显示金针菇蓝光受体FfWC-1和红光受体FfPhy同时存在于细胞质和细胞核中,其他3个光受体定位于细胞质中;跨膜区分析显示5个光受体都没有跨膜区。组织表达分析FfWC-1、FfWC-2和FfCry-DASH主要在成熟菌盖中表达,FfCpl和FfPhy主要在菌丝中表达。经蓝光处理和红光处理后,5个基因的表达模式也各不相同。在金针菇基因组中未发现有VIVID基因和绿光受体基因。本结果为进一步研究金针菇乃至食用菌的光生物学特征提供了分子理论基础。Four putative blue light receptor genes and one red light receptor gene were identified in the whole genome of Flammulina filiformis.The sequences of the five genes were analyzed by bioinformatics,including gene primary structure analysis,conservative structure analysis,subcellular localization prediction and transmembrane region analysis.The results of subcellular localization showed that the blue light receptor FfWC-1 and red light receptor FfPhy stimutaneously existed in the cytoplasm and nucleus,and the other three light receptors were localized in the cytoplasm.Transmembrane domain analysis showed that none of the five photoreceptors had transmembrane domain.Tissue expression indicated that FfWC-1,FfWC-2 and FfCry-DASH were mainly expressed in mature pileus,while FfCpl and FfPhy were mainly expressed in mycelium.After blue light and red light treatment,the expression patterns of the five genes were also different.VIVID gene and green light receptor gene were not found in the Flammulina filiformis genome.The results provide a molecular basis for further study on photobiological characteristics of Flammulina filiformis and even other edible fungi.
