摘 要:为了揭示咖啡活性成分咖啡豆醇(kahweol, KAH)对递增负荷运动大鼠能量代谢的影响及机制。本研究连续8周对递增负荷运动大鼠灌胃不同剂量(5, 10和20 mg·kg^(-1)·d^(-1))的咖啡豆醇。然后检测了大鼠的力竭游泳时间、肌肉和肝功能损伤指标(CK, ALT和AST)水平、能量代谢指标(肝糖原,肌糖原, LA, BUN和NEFA)水平,骨骼肌氧化应激指标(SOD, CAT和MDA)水平。通过RT-qPCR检测了骨骼肌中PGC-1α、TFAM、NRF1和ATP5B的mRNA水平。通过Western blot检测了骨骼肌中AMPKα的磷酸化水平。结果显示,不同剂量的咖啡豆醇升高了大鼠的力竭游泳时间,降低了血清CK、ALT和AST水平,升高了肝糖原和肌糖原含量,降低了血清LA和BUN水平,升高血清NEFA水平(P<0.05)。不同剂量的咖啡豆醇升高了骨骼肌SOD和CAT水平,降低了骨骼肌MDA水平(P<0.05)。不同剂量的咖啡豆醇升高了骨骼肌PGC-1α、TFAM、NRF1和ATP5B的mRNA相对表达量和p-AMPKα的蛋白相对表达量(P<0.05)。本研究表明咖啡活性成分咖啡豆醇有效提高了递增负荷运动大鼠的运动能力并改善了能量代谢,其机制可能与AMPK信号通路的激活有关。咖啡豆醇可能是一种改善运动功能的营养补充剂,具有较高开发潜力。The purpose of this study was to reveal the effect and mechanism of coffee active ingredient kahweol(KAH)on energy metabolism in rats with incremental load exercise.In this study,rats with incremental load exercise were given different doses(5,10 and 20 mg·kg^(-1)·d^(-1))of kahweol by stomach for 8 weeks.Then the exhaustive swimming time,the levels of muscle and liver function damage indexes(CK,ALT and AST),the levels of energy metabolism indexes(liver glycogen,muscle glycogen,LA,BUN and NEFA)and the oxidative stress indexes of skeletal muscle(SOD,CAT and MDA)were measured.The mRNA levels of PGC-1α,TFAM,NRF1 and ATP5B in skeletal muscle were detected by RT-qPCR.The phosphorylation level of AMPKαin skeletal muscle was detected by Western blot.The results showed that different doses of kahweol increased the exhaustive swimming time,decreased the levels of serum CK,ALT and AST,increased the contents of liver glycogen and muscle glycogen,decreased the levels of serum LA and BUN,and increased the level of serum NEFA(P<0.05).Different doses of kahweol increased the levels of SOD and CAT in skeletal muscle and decreased the level of MDA in skeletal muscle(P<0.05).Different doses of kahweol increased the relative mRNA expression of PGC-1α,TFAM,NRF1 and ATP5B and the relative expression of p-AMPKα protein in skeletal muscle(P<0.05).This study shows that kahweol,the active ingredient of coffee,can effectively improve the exercise ability and energy metabolism of rats with incremental load exercise,and its mechanism may be related to the activation of AMPK signal pathway.Kahweol may be a nutritional supplement to improve exercise function and has high development potential.
