摘 要:目的观察阿拉伯木聚糖(arabinoxylans,AX)对颈背部皮下注射鱼藤酮葵花油乳化液致帕金森病(Parkinson’s disease,PD)大鼠的行为学及肠道菌群的影响,探讨其对PD模型大鼠的作用及相关机制。方法选取40只SPF级健康雄性SD大鼠,空白组和假手术组各8只,其余24只造帕金森模型,并随机分为模型组、美多芭组、AX组,各组大鼠分别给予不同干预。干预14 d后进行行为学实验(悬挂实验)评估各组大鼠行为学变化;免疫组化检测大鼠黑质中酪氨酸羟化酶(tyrosine hydroxylase,TH)及α-突触核蛋白(α-SYNuclein,α-SYN)的表达;肠道菌群16S rRNA高通量测序检测各组大鼠肠道微生物的变化。结果AX组与模型组相比停留时间长(P<0.05);AX组与模型组相比α-SYN蛋白表达较低,TH蛋白表达含量较高;肠道菌群α多样性分析,与模型组相比,AX组和美多芭组ACE、Chao1、Shannon、Simpson指数升高,菌群多样性较丰富,差异有统计学意义(P<0.05);美多芭组与AX组相比,差异无统计学意义(P>0.05);线性判别分析重点菌群结果表明,AX能够升高厚壁菌门(Firmicutes)、梭状芽孢杆菌纲(Clostridia)、毛螺目(Lachnospirates)和毛螺科(Lachnospiraceae)相对丰度。结论AX可能通过调节肠道菌群改善PD大鼠症状进而达到治疗目的,或许未来可作为PD治疗方案的选择之一。Objective This study aimed to observe the effects of arabinoxylans(AX)on the behavior and intestinal flora of rats with Parkinson’s disease(PD)induced by subcutaneous injection of rotenone sunflower oil emulsion in the neck and back,and to explore the effect and related mechanism of AX on PD model rats.Methods A total of 40 SPF healthy male SD rats were selected,with 8 in the blank group and 8 in the sham group,and the remaining 24 rats were used to build Parkinson’s disease model and were randomly divided into the model group,the madopa group and the AX group.Each group was given different interventions.After 14 days of intervention,a behavioral experiment(suspension experiment)was performed to evaluate the behavioral changes of rats in each group.Immunohistochemistry was employed to detect the expression of tyrosine hydroxylase(TH)andα-synuclein(α-SYN)in the substantia nigra of rats.Intestinal microbiota 16s rDNA high-throughput sequencing was used to detect the changes of intestinal microbiota in each group.Results Compared with the model group,AX group had a longer residence time(P<0.05).Compared with the model group,AX group had lowerα-SYN protein expression and higher TH protein expression.Theαdiversity analysis of intestinal flora showed that,compared with the model group,the AX group and the madopa group had increased ACE,Chao1,Shannon and Simpson indicators,and richer diversity of intestinal flora,and the differences were statistically significant(P<0.05).There was no significant difference between the madopa group and AX group(P>0.05).Linear discriminant analysis showed that AX could increase the relative abundance of Firmicutes,Clostridia,Lachnospirates and Lachnospiraceae.Conclusion Arabinoxylan may improve the symptoms of PD rats by regulating intestinal flora and thus achieve the therapeutic purpose,which may be used as one of the options for PD treatment in the future.
