摘 要:探究了以植物乳杆菌N3117为活菌成分的益生菌发酵乳对二甲双胍治疗的糖尿病大鼠肠道菌群的影响。利用高脂饮食联合链脲佐菌素诱导2型糖尿病大鼠模型。将造模成功的大鼠随机分为糖尿病组(DM)、二甲双胍治疗组(Met)以及发酵乳和二甲双胍联合干预组(MLf);正常饮食的大鼠为对照组(Con)。每周记录大鼠体重及血糖水平。收集大鼠粪便,采用16S rRNA高通量测序对大鼠肠道菌群进行分析。与DM组大鼠相比,Met组空腹和餐后血糖显著降低(P<0.05),但仍显著高于Con组(P<0.05);与Con组相比,DM和Met组肠道菌群shannon指数和simpson指数均显著下降(P<0.05),而且厚壁菌门与拟杆菌门的比值显著降低(P<0.05);但与DM组相比,Met组寡源杆菌、棒状杆菌等肠道有害菌的丰度增加。与DM组相比,MLf组血糖水平显著降低(P<0.05),simpson指数显著升高(P<0.05),厚壁菌门与拟杆菌门的比值显著升高(P<0.05),乳酸杆菌、毛螺菌等有益菌的丰度增加,肠道菌群向Con组的组成靠近。结果表明联合干预能够在发挥降糖作用的同时,在一定程度上调节大鼠肠道菌群结构,缓解因糖尿病及二甲双胍治疗而导致的肠道菌群失调。To investigate the effects of probiotic fermented milk with Lactobacillus plantarum N3117 as living bacteria on intestinal microflora of diabetic rats treated with metformin were investigated.High-fat diet combined with streptozotocin was used to induce type 2 diabetes in rats.The rats were randomly divided into diabetic rats group(DM),metformin treatment group(Met),and combined fermented milk and metformin intervention group(MLf);rats with normal diet were the control group(Con).The body weight and blood glucose levels of rats were recorded weekly.Feces of rats were collected and the intestinal flora of rats was analyzed by 16S rRNA high-throughput sequencing.Compared with DM group,fasting and postprandial blood glucose in Met group was significantly decreased(P<0.05),but it was still significantly higher than that in Con group(P<0.05).The analysis of intestinal flora showed that compared with the Con group,the shannon index and simpson index of DM and Met groups markedly decreased(P<0.05).And the ratio of Firmicutes to Bacteroidetes decreased significantly between the DM and Met groups too(P<0.05).But compared with DM group,the abundance of intestinal harmful bacteria such as oligobacteria and Corynebacterium increased.Compared with DM group,the blood glucose level was significantly decreased(P<0.05)and simpson index were significantly increased in MLf group(P<0.05),and the ratio of Firmicutes to Bacteroidetes,and the abundance of beneficial bacteria such as Lactobacillus and Lachnospira was increased(P<0.05),the intestinal flora is close to the composition of Con group.The results showed that the combined intervention could not only play a hypoglycemic role,but also regulate the intestinal microflora structure of rats to a certain extent,and alleviate the intestinal microflora imbalance caused by diabetes and metformin treatment.
