摘 要:目的探讨缺血性脑卒中(IS)急性期外周血免疫特征和调控免疫反应的潜在机制。方法从GEO数据库中下载IS患者外周血的芯片测序数据,分析卒中组和对照组间免疫评分和免疫细胞的差异。筛选卒中组和对照组间的差异表达基因,并进行基因功能、通路富集分析。进行基因共表达调控网络分析(WGCNA),以筛选调控免疫特征的潜在分子标志物。结果与对照组相比,卒中患者的免疫评分显著升高,外周血巨噬细胞的丰度显著增加。基因功能富集分析显示,差异表达基因显著富集于免疫调节相关功能。结论FBXL13,DCLRE1C,ZIC3和ZNF528基因在卒中患者中的表达水平显著上调,并与免疫评分升高和巨噬细胞增多显著相关,可能成为评估卒中患者免疫特征的重要生物标志物以及潜在的干预靶点。Objective To investigate the immune characteristics of peripheral blood in acute phase of ischemic stroke and the potential mechanism of regulating immune response.Methods The microarray sequencing data of peripheral blood of patients with ischemic stroke were downloaded from the Gene Expression Omnibus(GEO)database.The differences in ImmuneScore and immune cells between the stroke group and the control group were analyzed.The differentially expressed genes between the stroke group and the control group were screened,and gene function and pathway enrichment analysis were performed.Weighted correlation network analysis(WGCNA)was performed to screen for potential molecular markers regulating immune characteristics.Results Compared with the controls,the immune scores of stroke patients were significantly higher,and the abundance of macrophages was also significantly increased.Gene function enrichment analysis showed that differentially expressed genes were significantly enriched in immunomodulatory functions.Conclusion The expression levels of FBXL13,DCLRE1C,ZIC3 and ZNF528 genes were significantly up-regulated in stroke patients,and were significantly correlated with the increase of Immune Score and macrophages,which may be used as important biomarkers to evaluate the immune characteristics of stroke patients and as potential therapeutic targets.
