摘  要:为了探究广叶绣球菌多糖(SLPs)对染铅小鼠骨骼钙代谢的影响,将小鼠随机分为对照组、染铅组和低、中、高剂量SLPs组,饲养8周后采样,采用电感耦合等离子发射光谱仪测定骨骼中二价金属元素的含量;酶联免疫吸附法检测骨骼中甲状旁腺激素(PTH)、骨钙素(BGP)、降钙素(CT)含量和碱性磷酸酶(AKP)活性;HE染色观察小鼠股骨组织形态;实时荧光定量PCR检测骨骼中钙代谢相关基因的表达量。结果表明,与对照组相比,染铅组骨骼中Pb^(2+)、PTH含量和PMCA1b mRNA表达量极显著升高,Ca^(2+)、Fe^(2+)、Cu^(2+)、Mn^(2+)、BGP、CT含量以及AKP活性和TRPV5、TRPV6、NCX1 mRNA表达量极显著下降,染铅组的骨小梁分布紊乱,骨小梁边缘出现陷窝。与染铅组相比,中剂量SLPs组Fe^(2+)含量极显著上升,高剂量SLPs组骨骼中Pb^(2+)、PTH含量、PMCA1b mRNA表达量显著下降,Ca^(2+)、CT含量、AKP活性、TRPV5、TRPV6 mRNA表达量显著上升,高剂量SLPs组的骨小梁分布较均匀,骨小梁边缘陷窝较少。SLPs可以促进机体对铅的排出,改善铅致骨骼中钙铁元素失衡,降低骨骼中PTH水平,提高骨骼中CT含量和AKP活性,缓解铅对骨骼组织形态结构的损伤,调控钙代谢相关基因的表达,进而对铅致小鼠骨骼钙代谢紊乱产生调节作用。The study was designed to investigate the effects of Sparassis latifolia polysaccharides(SLPs)on skeletal calcium metabolism in lead-contaminated mice.The mice were randomly divided into control group,lead group,low-dose SLPs group,medium-dose SLPs group,and high-dose SLPs group.After 8 weeks of feeding,the bones were collected to determine the contents of divalent metal elements by inductively coupled plasma emission spectrometer,the contents of parathyroid hormone(PTH),bone glaprotein(BGP),calcitonin(CT),and the activity of alkaline phosphatase(AKP)by enzyme-linked immunosorbent assay,observe morphology of mouse femur tissue by HE staining,and detect the expression levels of calcium metabolism related genes in bones by real-time fluorescence quantitative polymerase chain reaction.The results showed that compared with the control group,the contents of Pb^(2+),PTH and the expression levels of PMCA1b mRNA in bones in the lead group were significantly increased,while the contents of Ca^(2+),Fe^(2+),Cu^(2+),Mn^(2+),BGP,and CT,the AKP activity,and the expression levels of TRPV5,TRPV6,and NCX1 mRNA were significantly decreased.The distribution of bone trabecula was disordered and lacunae appeared at the margin of bone trabecula in lead group.Compared with the lead group,the Fe^(2+)content was significantly increased in the medium-dose SLPs group.The contents of Pb^(2+),PTH and the expression levels of PMCA1b mRNA in high-dose SLPs group were significantly decreased,while the contents of Ca^(2+)and CT,AKP activity,and the expression levels of TRPV5 and TRPV6 mRNA were significantly increased.In the high-dose SLPs group,the bone trabecular was more evenly distributed and the margin lacunae of bone trabeculae was less.It indicated that SLPs could improve skeletal imbalance of calcium and iron elements induced by lead in mice by promoting the excretion of lead,reducing the PTH level in bones,increasing the CT content and the AKP activity in bones,alleviate the damage of lead on morphological structures in bone tissues,and
