摘  要:目的探讨表没食子儿茶素没食子酸酯((-)-epigallocatechin gallate,EGCG)是否影响了辐射对食管癌Eca-109细胞的作用及活性氧(reactive oxygen species,ROS)的变化。方法将Eca-109细胞分为4组:对照组(control组)、辐射组(2 Gy,24 h)、EGCG组(20μmol/L,24 h)和辐射+EGCG组。流式细胞仪检测细胞周期、细胞凋亡比例和ROS变化。Western blot检测Caspase-3和Bcl-2蛋白表达。克隆形成实验检测放射敏感性指标平均致死剂量(D 0)、准阈剂量(D q)及靶数值变化。结果与control组和EGCG组相比,辐射组与辐射+EGCG组G2/M期细胞比例降低(P<0.05);其余组间差异无统计学意义(P>0.05)。与control组和EGCG组相比,辐射组与辐射+EGCG组细胞凋亡率和Caspase-3蛋白表达增多(P<0.05);与辐射组相比,辐射+EGCG组细胞凋亡率和Caspase-3蛋白表达增多(P<0.05);其余组间差异无统计学意义(P>0.05)。与control组和EGCG组相比,辐射组Bcl-2蛋白表达降低(P<0.05),其余组间差异无统计学意义(P>0.05)。与control组相比,辐射组ROS含量增多(P<0.05),EGCG组ROS含量减少(P<0.05);与辐射组相比,EGCG组与辐射+EGCG组ROS含量减少(P<0.05);其余组间差异无统计学意义(P>0.05)。与control组相比,EGCG组D q和靶数值降低(P<0.05),D 0值无明显差异(P>0.05)。结论EGCG没有降低辐射对Eca-109细胞的抗肿瘤作用,但降低了辐射诱导的ROS生成。Objective To investigate the effect of EGCG on the antitumor role of radiation and the change of ROS in esophageal cancer Eca-109 cells.Methods Eca-109 cells were divided into four groups:control group,radiation group(2 Gy,24 h),EGCG group(20μmol/L,24 h)and radiation+EGCG group.The changes of cell cycle,apoptosis rate and ROS were detected by flow cytometry.Western blot assay was used to detect the protein expression of Caspase-3 and Bcl-2.Colony formation assay was performed to analyze the changes of mean lethal dose(D 0),quasithreshold dose(D q)and extrapolation number of radiosensitivity.Results Compared with control group and EGCG group,the proportion of cells in G2/M phase was decreased significantly in radiation group and radiation+EGCG group(P<0.05),and there was no significant difference between the other groups(P>0.05).Compared with control group and EGCG group,the apoptosis rate and the expression of Caspase-3 protein were increased significantly in radiation group and radiation+EGCG group(P<0.05).Compared with radiation group,the apoptosis rate and the expression of Caspase-3 protein were enhanced significantly in radiation+EGCG group(P<0.05),but there was no significant difference between the other groups(P>0.05).Compared with control group and EGCG group,the protein expression of Bcl-2 in radiation group was decreased significantly(P<0.05),but there was no significant difference between the other groups(P>0.05).Compared with control group,ROS was increased significantly in radiation group(P<0.05),and decreased in EGCG group(P<0.05).Compared with radiation group,ROS was decreased significantly in EGCG group and radiation+EGCG group(P<0.05),but there was no significant difference between the other groups(P>0.05).Compared with control group,D q and extrapolation number in EGCG group were decreased significantly(P<0.05).There was no significant difference in D 0 value between EGCG group and control group(P>0.05).Conclusion EGCG can not affect the antitumor effect of radiation on Eca-109 cells,but reduce
