摘 要:采用超声波法,以余甘子果为原料、SOD酶比活力为指标,考查液料比、磷酸缓冲液pH、超声时间、提取温度对余甘子SOD提取率的影响,并根据单因素试验结果及响应面Box-Behnken中心拟合原理,以SOD酶比活力为响应值,进行4因素3水平试验设计,建立响应面优化超声波法提取余甘子SOD工艺的多元回归方程;同时采用DPPH自由基和羟基自由基清除法对提取出的余甘子SOD粗酶液进行了抗氧化性研究.实验结果表明:响应面优化超声波法提取余甘子SOD最佳工艺为液料比5∶1(mL/g)、磷酸缓冲液pH值7.8、超声时间19 min、温度50℃,在此工艺下进行3次平行验证得到SOD酶比活值为(557.22±9.36)U/g(FW)·h,与模型预测值573.36 U/g(FW)·h误差为2.90%,误差值小于5%说明模型可靠性高;余甘子的SOD粗酶液对DPPH自由基清除率最高为97.41%,羟基自由基的清除率最高也可达到93.36%,且抗氧化性优于同等浓度的VC;优化后的余甘子中SOD的提取工艺稳定可靠,且SOD粗酶液具有良好的抗氧化活性.Using Phyllanthus emblica as raw material and SOD enzyme specific activity as index,the effects of liquid to material ratio,phosphate buffer pH,ultrasonic time and extraction temperature on the extraction rate of Phyllanthus emblica SOD were investigated by ultrasonic method.According to the results of single factor test and the Box-Behnken center fitting principle of response surface,taking the specific activity of SOD enzyme as the response value,the experimental design of 4 factors and 3 levels was carried out,and the multiple regression equation of ultrasonic method for extracting SOD from Phyllanthus emblica was established,and the antioxidant activity of the crude enzyme solution of Phyllanthus emblica SOD was studied by DPPH radical and hydroxyl radical scavenging method.The experimental results show that the best process for extracting SOD from Phyllanthus emblica by response surface optimization ultrasonic method is as follows:liquid to material ratio 5∶1(mL/g),pH value of phosphate buffer 7.8,ultrasonic time 19min,temperature 50℃.After three times of parallel verification under this process,the specific activity of SOD is(557.22±9.36)U max g(FW)h,and the error is 2.90%compared with the predicted value of 573.36 U max g(FW)h.The error value less than 5%indicates that the model has high reliability.The highest scavenging rate of DPPH radical and hydroxyl radical was 97.41%and 93.36%respectively,and the antioxidant activity was better than that of the same concentration of VC.The optimized SOD extraction technology was stable and reliable,and the SOD crude enzyme liquid had good antioxidant activity.
