摘 要:【目的】为探究贮藏温度对南京椴种子耐贮性的影响,揭示不同贮藏温度条件下种子发芽率和生理生化指标的变化规律,旨在探寻种子适宜贮藏温度,为南京椴种子贮藏提供科学依据。【方法】以含水量为9.80%的南京椴种子为试验材料,存放在室温(23~27℃)、4、-5和-40℃等4种温度条件下开展种子贮藏,对贮藏0、180、360、540、720 d的种子分别测定种子绝对发芽率和相对电导率、SOD活性、POD活性、O_(2)^(−)·含量、OH·产生能力与MDA含量等生理生化指标。【结果】贮藏温度和贮藏时间协同影响南京椴种子活力。随着贮藏时间的延长,各贮藏温度条件的种子发芽率均下降;随着贮藏温度的下降,种子发芽率显著上升;至720 d贮藏试验结束时,在室温、4、-5和-40℃贮藏条件下的种子绝对发芽率分别为43%、49%、60%、62%。贮藏时间延长导致种子发生不同程度的劣变,不同贮藏温度条件下的种子相对电导率均呈上升趋势;在贮藏前期,种子SOD和POD活性升高,后期均有所下降;种子O_(2)^(−)·含量、OH·产生能力和MDA含量整体均呈逐渐上升的变化趋势。【结论】室温贮藏180 d时,南京椴种子仍可保持较高的活力水平;但随着贮藏时间的继续延长,降低贮藏温度有利于提高种子耐贮性,-5和-40℃条件下贮藏的种子自由基积累量低于室温与4℃条件下贮藏的种子且种子抗氧化能力较强;综合比较控温条件和节约能耗,建议选择-5℃作为南京椴种子贮藏的环境温度。【Objective】In order to explore the effect of storage temperature on the storability of Tilia miqueliana seeds,and reveal the changes of seed germination rate and physiological and biochemical indexes under different storage temperature conditions,this paper aimed to explore the suitable storage temperature of seeds and provide a scientific basis for the storage of Tilia miqueliana seeds.【Method】Tilia miqueliana seeds with a water content of 9.80%were used as the test material and stored at room temperature(23-27℃),4,-5 and-40℃for seed storage,and physiological and biochemical indexes such as absolute germination rate,relative conductivity,SOD activity,POD activity,O_(2)^(−)·content,OH·production capacity and MDA content were determined for seeds stored at 0 d,180 d,360 d,540 d and 720 d.【Result】Storage temperature and storage time synergistically affected the vitality of Tilia miqueliana seeds.With the extension of storage time,the germination rate of seeds under various storage temperature conditions decreased.With the decrease of storage temperature,the germination rate of seeds increased significantly.By the end of the 720 d storage test,the absolute germination rates of seeds at room temperature,4,-5 and-40℃storage conditions were 43%,49%,60%and 62%,respectively.The prolongation of storage time led to different degrees of seed deterioration,and the relative conductivity of seeds under different storage temperature conditions showed an increasing trend.In the early stage of storage,the activity of seed SOD and POD increased,and decreased in the later stage.The overall trend of seed O_(2)^(−)·content,OH·production capacity and MDA content showed an upward trend.【Conclusion】When stored at room temperature for 180 days,Tilia miqueliana seeds could still maintain a high level of vitality.However,with the continuous extension of storage time,reducing the storage temperature is conducive to improving the storage tolerance of seeds,and the accumulation of free radicals in seeds stored at
