青楷槭茎皮乙醇提取工艺优化和抗氧化性研究

摘  要:为优化青楷槭茎皮乙醇提取的工艺条件并研究其抗氧化能力、进一步分离青楷槭茎皮生物活性成分、丰富槭树科的利用价值,利用正交试验对青楷槭茎皮乙醇提取条件进行优化,并通过测定最优条件提取物的DPPH·、ABTS^(+)·两种自由基的清除率以评价青楷槭茎皮醇提物的抗氧化作用。结果表明,青楷槭茎皮乙醇提取物最优提取条件为料液比1∶10(g/mL)、乙醇体积分数60%、浸泡时间6 h,在最优条件下青楷槭茎皮醇提物的总酚含量为577.8μg/mg,并且当青楷槭茎皮醇提物浓度达到500μg/mL时,DPPH·清除率为89.26%,ABTS^(+)·清除率为92.98%。通过与BHA对比发现,其抗氧化作用随浓度升高而增强,呈现出剂量效应关系,表明青楷槭茎皮醇提物具有良好抗氧化能力。In order to optimize the extraction process of ethanol from stem peel of Acer cymbium and study its antioxidant capacity,to further isolate the biological active components from the stem bark of Acer cydendrum,and to enrich the utilization value of Aceraceae,orthogonal experiment was used to optimize the ethanol extraction conditions of Acer cydendrum stem peel,and the antioxidant activity of the ethanol extract from Acer cydendrum stem peel was evaluated by measuring the scavenging rates of DPPH·and ABTS^(+)·of the optimal extract.The results showed that the optimal extraction conditions were solid-liquid ratio of 1∶10(g/mL),ethanol concentration of 60%and soaking time of 6 h.Under the optimal conditions,the total phenolic content of the ethanol extract was 577.8μg/mg.When the ethanol concentration reached 500μg/mL,the DPPH·scavenging rate was 89.26%,and the ABTS^(+)·scavenging rate was 92.98%.Compared with BHA,the antioxidant activity increased with increasing concentration,showing a dose-effect relationship,indicating that the ethanol extracts from stem bark of Acer cydendrum had good antioxidant capacity.
