摘 要:建立大孔树脂纯化蒲菜黄酮化合物的工艺条件,并讨论其对小鼠运动耐力的影响。根据单因素试验结果,确定AB-8型大孔树脂纯化蒲菜总黄酮的最佳条件为上样浓度3.0 mg/mL、上样液pH 4.0、上样流速与洗脱流速分别为2.0 mL/min和1.0 mL/min,洗脱液乙醇体积分数为65%,上样液体积与洗脱液体积分别为70 mL与80 mL。纯化后的产物中黄酮化合物的纯度由18.3%提高至64.5%,约为纯化前的3.5倍。与空白对照组比较,低、高剂量纯化物组小鼠的负重游泳时间均有明显延长(P<0.05,P<0.01),血清中乳酸与尿素氮浓度均显著下降(P<0.05,P<0.01),同时肝脏与骨骼肌组织中丙二醛的生成量明显减少(P<0.05,P<0.01),并可增强体内超氧化物歧化酶的活力(P<0.05,P<0.01),从而可知蒲菜黄酮化合物能够提高机体的运动耐力。The purification process condition of total flavonoids from Typha latifolia using macroporous resin was established,and its effect on exercise endurance of mice was discussed.According to the results of single factor test,the optimum purification process conditions were confirmed that the concentration of loading solution was 3.0 mg/mL,and pH was 4.0,the volume fraction of ethanol acted as eluent was 65%,flow rate of loading sample and eluent were 2.0 mL/min and 1.0 mL/min,respectively.Under these conditions,the purity of total flavonoids in purified product was increased from 18.3%to 64.5%,which was about 3.5 times higher than that before purification.Compared with blank control group,the weight-loading swimming time of mice in the purified product group at low and higher dose were obviously prolonged(P<0.05,P<0.01),and accumulation of blood lactic acid and urea nitrogen in serum were significantly decreased(P<0.05,P<0.01).Meanwhile,the generation of malonaldehyde in tissue of liver and skeletal muscle were decreased(P<0.05,P<0.01)in the purified product group at low and higher dose and they could strengthen activity of superoxide dismutase(P<0.05,P<0.01).Therefore,total flavonoids from Typha latifolia could improve the exercise endurance of body.
