青岛市主干道3种植物叶表形态及叶际微生物群落结构研究

摘  要:为探究青岛市主干道3种植物叶表面颗粒物形态特征及微生物群落结构,选择冬青、女贞和樱花作为供试树种,利用环境扫描电镜观察植物叶片微观结构和微生物微形态变化,采用16S rRNA高通量测序分析叶际细菌群落多样性与丰富度。结果显示:3种植物叶片滞尘能力冬青>女贞>樱花,冬青、女贞在夏冬季节滞尘能力强,樱花在夏秋季节滞尘能力强;扫描电镜显示植物叶表面存在真菌、真菌孢子、丝状真菌;高通量测序结果显示植物群落多样性无显著差异,叶际细菌群落丰富度呈现冬青>女贞>樱花,冬青叶际细菌群落丰富度秋季最低,女贞和樱花植物叶际细菌群落丰富度夏季最低;3种植物叶表面主要优势菌门为Proteobacteria,Streptophyta和Bacteroidetes,主要优势菌纲为Gammaproteobacteria,Magnoliopsida,Betaproteobacteria,Alphaproteobacteria。研究表明:城市主干道3种植物中冬青滞尘能力最强,可以为选择城市园林树种提供指导;相对于季节变化,叶表面颗粒物和微生物群落结构更易受植物类型的影响。In order to explore the morphological characteristics of the particulate matter on leaf surface and the microbial community structure of three kinds of plants on the main roads of Qingdao,Ilex chinensis Sims,Ligustrum compactum and Prunus serrulata were selected as the test trees,the changes of the plant leaf microstructure and the microbial micro morphology were observed by using environmental scanning electron microscope,and the 16S rRNA high-throughput sequencing was used to analyze the diversity and richness of bacterial community in the leaf.The results showed that the dust retention ability of the leaves of the three plants was I.chinensis Sims>L.compactum>P.serrulate.I.chinensis Sims and L.compactum had strong dust retention ability in summer and winter,and P.serrulata had strong dust retention ability in summer and autumn.Scanning electron microscope showed that there were fungi,fungal spores and filamentous fungi on the surface of plant leaves.The results of high-throughput sequencing showed that there was no significant difference in plant community diversity.The abundance of foliar bacterial community was I.chinensis Sims>L.compactum>P.serrulata.The abundance of foliar bacterial community of I.chinensis Sims was the lowest in autumn,and that of L.compactum and P.serrulata was the lowest in summer.The main dominant bacteria classes on the leaf surface of the three plants are Proteobacteria,Streptophyta and Bacteroidetes,and the main dominant bacteria phyla are Gammaproteobacteria,Magnoliopsida,Betaproteobacteria and Alphaproteobacteria.The results showed that among the three plants on urban trunk roads,I.chinensis Sims has the strongest dust retention ability,which can provide guidance for the selection of urban garden tree species.Compared with seasonal changes,leaf surface particulate matter and microbial community structure are more vulnerable to plant types.
