摘  要:通过蛋白质组学筛选和研究不同花性山葡萄授粉前后雌蕊的差异蛋白质变化,揭示山葡萄授粉受精过程的内在分子调控机制。利用iTRAQ技术筛选与鉴定授粉受精前后山葡萄两性花‘双丰’与雌能花‘左山一’雌蕊的差异蛋白。结果表明,COG注释的翻译后修饰、核糖体结构、能量产生、物质运输与代谢类的蛋白数量最多;共鉴定到351个差异表达蛋白,‘双丰’花后上调91个,下调58个;‘左山一’花后上调127个,下调75个;11条显著性富集的KEGG通路,主要包括:核糖体、光合作用、光合作用—天线蛋白、N-聚糖生物合成、内质网蛋白加工、过氧物酶体及代谢。差异表达蛋白主要涉及核糖体、光合作用、能量代谢与转换、花粉管生长,前三种蛋白的表达量均发生显著变化,花粉管生长蛋白只在‘双丰’中显著下调表达,‘左山一’授粉前后的雌蕊中均未检测到此类蛋白,可能与雄蕊发育存在一定关系。To investigate the molecular mechanisms of the development of Vitis amurensis pistil after pollination,protein abundance changes were studied by using approach of proteomics to screen the different proteins related to the flower sexuality and fertilization process of Vitis amurensis.The iTRAQ technique was used to screen and identify the DEPs(differentially expressed proteins)in the pistils of Vitis amurensis'Shuangfeng'(hermaphrodite flower cultivar)and'Zuoshan1'(male sterile cultivar)before and after pollination.The results showed that COG annotations including posttranslational modification,ribosome structure,energy generation,transportation and metabolism are the most.A total of 351 DEPs were identified in the paired comparison of pistil after pollination,including 127 up-regulated proteins and 75 down-regulated proteins after anthesis of'Zuoshan1',91 up-regulated proteins and 58 down-regulated proteins after anthesis of'Shuangfeng'.And 11 significant enriched KEGG pathways were identified by these DEPs,including ribosome,photosynthesis,photosynthesis-antenna protein,N-glycan biosynthesis,protein processing in endoplasmic reticulum,peroxisome and metabolic pathways.DEPs mainly involved ribosome,photosynthesis,energy production and conversion and pollen tube growth.The expression of former three proteins was significantly changed in both'Zuoshan1'and'Shuangfeng',but the pollen tube growth proteins which were not detected in'Zuoshan1'pistil after pollination were down-regulated in'Shuangfeng'.There might be some relationships between the development of stamen and pistil and it needed further study.
