摘 要:通过研究以无花果叶为基质培养枯草芽孢杆菌对其抑菌能力、抗氧化能力的影响,为提高无花果叶的综合利用率及深度利用枯草芽孢杆菌提供思路。本研究采用以无花果叶为基质培养枯草芽孢杆菌(试验组),同无花果叶研磨液(对照Ⅰ组)与营养培养基(对照Ⅱ组)培养枯草芽孢杆菌形成对比,采用滤纸片法及抗氧化试剂盒测定三组发酵液的抑菌作用以及抗氧化活性,利用代谢组学技术检测代谢物,分析使其产生差异的代谢物。结果表明:营养培养基枯草芽孢杆菌发酵液抑菌圈直径为19.3 mm,以无花果叶为基质培养枯草芽孢杆菌的发酵液抑菌圈直径为24.3 mm。以无花果叶为基质培养的枯草芽孢杆菌,发酵液清除DPPH自由基、超氧阴离子自由基、羟自由基能力明显强于营养培养基培养枯草芽孢杆菌。通过代谢组学技术,试验组筛选出98种差异代谢物,对照Ⅱ组中53种差异代谢物。对这些代谢物进行分类发现,脂质类含量最高,生物碱类、抗生素类、萜类、黄酮类、酚类、部分氨基酸类代谢物含量较高,其中,试验组中有59种相关物质,对照Ⅱ组中有28种,这些物质可能对提高枯草芽孢杆菌抑菌能力、抗氧化能力发挥重要作用。To improve the comprehensive utilization of fig leaves as well as to provide ideas for the deep utilization of Bacillus subtilis,the effects of Bacillus subtilis culture in the fermentation of fig leaves on its biological activity were studied.In the experiments,fig leaves were used as a substrate to cultivate Bacillus subtilis(experimental group),which compared with fig leaves grinding liquid(control groupⅠ)and ordinary culture medium(control groupⅡ).The antibacterial effects and antioxidant activities of the three groups of fermentation broths were determined by filter paper method and antioxidant kits.The metabolomics analysis was used to detect metabolites and analyze the different metabolites.The results showed that the diameter of inhibition zone of Bacillus subtilis cultured in ordinary medium was 19.3 mm,and the diameter of inhibition zone of Bacillus subtilis cultured in fig leaves was 24.3 mm.The scavenging ability of Bacillus subtilis cultured in fig leaf on DPPH radical,superoxide anion radical and hydroxyl radical was significantly stronger than that of Bacillus subtilis cultured in ordinary medium.By metabolomics technology,98 differential metabolites were detected in the experimental group,and 53 differential metabolites were detected in the control groupⅡ.By classifying these metabolites,it was found that lipids had the highest content,followed by alkaloids,antibiotics,terpenoids,flavonoids,phenols,and some amino acid metabolites.Among them,there were 59 related substances in the experimental group and 28 in the control groupⅡ.These substances may play an important role in improving the antibacterial ability and antioxidant ability of Bacillus subtilis.
