摘 要:Micrococcus luteus ML是从长期使用毒死蜱的农田土壤中分离出的一株能够降解毒死蜱及其高毒残留物3,5,6-三氯-2-吡啶酚(TCP)的菌株。探究菌株ML降解机制,挖掘新型TCP代谢基因和特异代谢功能。采用Illumina Novaseq PE150和Oxford Nanopore ONT相结合的测序技术对菌株ML进行全基因组测序。该菌株有52个编码基因参与了异源化合物的代谢,共有15种酶参与了TCP结构类似物,如氯苯甲酸、氨基苯甲酸盐、甲苯、萘、氯代烷烃和氯代烯烃的降解。初步推测羧甲基丁内酯酶和酰胺酶分别参与了菌株ML对TCP的脱氯和脱氯产物吡啶环中氮转化为氨的反应,这为探究Micrococcus luteus ML功能基因组学研究及对TCP的降解特性提供了数据支持。Micrococcus luteus ML,capable of degrading chlorpyrifos and its highly toxic residue 3,5,6-trichloro-2-pyridinol(TCP),was isolated from soil where chlorpyrifos has been used for a long time.To investigate the degradation mechanism and mining novel TCP metabolic genes and specific metabolic functions of strain ML.Whole genome sequencing of strain ML with a combined sequencing technology of Illumina Novaseq PE150 and Oxford Nanopore ONT was performed.There are 52 coding genes relating to xenobiotics biodegradation and metabolism in strain ML,and a total of 15 enzymes might be involved in the degradation of TCP structural analogs,such as chlorobenzoic acid,aminobenzoates,toluene,naphthalene,chloroalkane and chloroalkene.It was speculated that carboxymethylenebutenolidase and amidase were involved in the dechlorination of TCP and the conversion of nitrogen to ammonia in pyridine ring of dechlori-nation metabolites,respectively.This article provides data for exploring the functional genomics of Micrococcus luteus ML and its degradation characteristics to TCP.
