摘  要:喷雾干燥制备益生菌制剂具有成本低、处理量大且可连续化生产的优势,但由于高温会损伤菌体活性因此喷雾干燥的应用受限。利用低温喷雾干燥技术降低热损伤,提高菌体存活率。通过单因素试验和响应面中心组合设计联用的方法优化工艺条件,以制备植物乳植杆菌P-8活性菌粉,并探究其菌株形态变化、贮藏稳定性以及在动态体外仿生胃肠道消化模型中的耐受性。植物乳植杆菌P-8在进料速度9 mL/min、干燥温度75℃、雾化压强为0.3 MPa时,存活率最高为81.50%,且菌粉颗粒形态完整。在动态体外仿生胃肠道消化模型内连续消化4 h后,表现出良好的耐受特性。菌粉4和25℃贮藏35 d后存活率分别为92.18%和87.49%。低温喷雾干燥技术可用于植物乳植杆菌P-8高活性菌粉的制备,优化后的工艺参数可以为植物乳植杆菌P-8中试制备提供参考。The preparation of probiotic preparations by spray drying has the advantages of low cost,large processing capacity and continuous production.However,its application is limited by the high temperature damage to the activity of the bacteria.To reduce the thermal damage and improve the survival rate of the bacteria,low temperature spray drying was applied in this study.Through single factor experiment and response surface center combination design,the optimum conditions to prepare Lactiplantibacillus plantarum P-8 active bacteria powder were obtained.The morphological changes and storage stability of strain were concerned.Tolerance in a dynamic extracorporeal bionic gastrointestinal digestive model was explored.The highest survival rate of L.plantarum P-8 was 81.50%at the feed rate of 9 mL/min,drying temperature of 75℃and atomization pressure of 0.3 MPa,while the form of bacteria powder particles remained complete.Good tolerance showed after continuous digestion for 4 h in a dynamic in vitro bionic gastrointestinal digestion model.The survival rates of the bacterial powder were 92.18%and 87.49%after 35 days of storage at 4 and 25℃,respectively.The low-temperature spray drying technology can be used for the preparation of highly active bacterial powder of L.plantarum P-8.The optimized process parameters provide a reference for the pilot preparation of L.plantarum P-8.
