低次烟叶浸膏的提取及其抗氧化活性

摘　　要：采用乙醇提取法、酶法、索氏提取法从低次烟叶中制备浸膏,首先通过气相色谱-质谱联用(GC-MS)分析了3种方法提取得到的浸膏中挥发性成分差异;其次通过测定DPPH·、ABTS+·、铁离子还原能力3个抗氧化指标来评估其体外抗氧化活性;并通过相关性分析研究了低次烟叶浸膏中的抗氧化活性与总酚、总黄酮、总糖含量的相关性;最后将3种方法提取得到的浸膏添加在卷烟中进行感官评价。结果表明,3种提取方法中酶法得率最高;经GC-MS分析,3种浸膏共检出了70种组分;3种浸膏均表现出体外抗氧化活性,醇提浸膏的清除效果最佳;在总酚、总黄酮、总糖的测定中,醇提浸膏中的含量更高;通过相关性分析发现,酚类和黄酮类的含量与抗氧化活性有密切关系;3种浸膏对卷烟都有明显的增甜增香和降低刺激性的效果。In this work,extracts were prepared from low-grade tobacco leaves by using ethanol extraction,enzymatic method and Soxhlet extraction.Firstly,the differences of volatile components from the extracts by these three methods were analyzed by gas chromatography-mass spectrometry(GC-MS).Secondly,three antioxidant indexes,DPPH·,ABTS+·and ferric reduction capacity,were measured to evaluate the antioxidant activity in vitro.The correlation between the antioxidant activity of the extracts from low-grade tobacco and the contents of total phenols,total flavonoids,and total sugars were studied by correlation analysis.Finally,the extracts were added to the cigarette for sensory evaluation.The results showed that enzymatic method had the highest yield among the three extraction methods.By GC-MS analysis,a total of 70 components were detected in the three extracts.All the three extracts showed antioxidant activity in vitro,and the ethanol extracts had the best scavenging effect.The alcohol extract had higher contents of total phenols,total flavonoids,and total sugars.The correlation analysis showed that the contents of phenols and flavonoids were closely related to the antioxidant activity.Three kinds of extracts have obvious effect of sweetening and reducing the irritant.
