摘 要:为了优化毛果算盘子粗提物提取条件,研究其体外抑菌效果,试验采用水提法提取毛果算盘子粗提物,并利用正交试验优化提取条件,以纸片法和微量肉汤释法对抑菌活性进行测定。结果表明:毛果算盘子粗提物最佳提取条件为料液比1∶30,100℃提取0.5 h、提取2次;其对金黄色葡萄球菌、耐头孢喹肟金黄色葡萄球菌、铜绿假单胞菌、鼠伤寒沙门氏菌的抑菌圈直径分别为(25.00±0.82)mm、(30.70±0.94)mm、(14.00±0.47)mm、(11.70±0.82)mm;最小抑菌浓度分别为1.56 mg/mL、1.56 mg/mL、3.125 mg/mL、3.125 mg/mL;最小杀菌浓度分别为1.56 mg/mL、3.125 mg/mL、3.125 mg/mL、6.25 mg/mL。说明毛果算盘子粗提物对四种试验菌株的抑制作用不同,对耐头孢喹肟金黄色葡萄球菌的抑制作用最强,对鼠伤寒沙门氏菌的抑制作用最弱。In order to optimize the extraction conditions of crude extract of Glochidion eriocarpum,and to study its antibacterial effect in vitro,in this experiment,the crude extract of Glochidion eriocarpum was extracted by aqueous extraction method,and the extraction conditions were optimized by orthogonal test;the antibacterial activity was determined by Kirby-Bauer method and micro-broth dilution method.The results showed that the optimal extraction conditions for crude extract of Glochidion eriocarpum were a material-liquid ratio of 1∶30,100 ℃ for 0.5 h and 2 times by extraction.The diameters of the inhibition circle against Staphylococcus aureus,cefquinoxime-resistant Staphylococcus aureus,Pseudomonas aeruginosa and Salmonella typhimurium were(25.00±0.82) mm,(30.70±0.94) mm,(14.00±0.47) mm,and(11.70±0.82) mm,respectively.The minimum inhibitory concentrations were 1.56 mg/mL,1.56 mg/mL,3.125 mg/mL and 3.125 mg/mL,respectively.The minimum bactericidal concentrations were 1.56 mg/mL,3.125 mg/mL,3.125 mg/mL and 6.25 mg/mL,respectively.The results suggested that the crude extract of Glochidion eriocarpum had different inhibitory effect on the four test strains;the inhibitory effect on cefquinoxime-resistant Staphylococcus aureus was the strongest,and the inhibitory effect on Salmonella typhimurium was the weakest.
