摘  要:目的:优化复合酶法提取莪术多糖的工艺,并研究其体外抗氧化活性。方法:以莪术多糖得率为指标,利用单因素试验和响应面试验探究莪术多糖提取的最优工艺,因素包括酶添加量、酶解时间、酶解温度及液料比。以DPPH自由基、羟自由基和超氧阴离子自由基清除率大小为指标考察莪术多糖的体外抗氧化活性。结果:复合酶由纤维素酶、果胶酶和木瓜蛋白酶按质量比1∶1∶1组成。复合酶提取莪术多糖最佳工艺为酶添加量3.5%、酶解时间100 min、酶解温度55℃、液料比(25∶1)mL/g、酶解pH5.0,多糖得率为5.18%,回归模型预测的莪术多糖得率为5.21%,两者误差小于5%。莪术多糖对DPPH自由基、羟自由基和超氧阴离子自由基的半数抑制浓度依次为1.169、1.284、1.076 mg/mL,表明其具有较强的自由基清除能力。结论:复合酶法可显著提高莪术多糖得率,工艺简便可行,获得的莪术多糖具有体外抗氧化活性。To optimize the compound enzyme extraction process of polysaccharide from Curcuma zedoary and study its antioxidant activity in vitro,the yield of polysaccharide from Curcuma zedoary was used as experimental index.Single factor test and response surface methodology were used to optimize the extraction process of polysaccharide from Curcuma zedoary.The factors included the amount of enzyme,enzymatic hydrolysis time,temperature and liquid-to-material ratio.DPPH radical,hydroxyl radical and superoxide anion radical scavenging rates were used as indexes to investigate antioxidant activities of polysaccharides in vitro.The results showed that the compound enzyme was composed of cellulase,pectinase and papain in a mass ratio of 1∶1∶1.The optimal extraction conditions were:enzyme 3.5%,enzymatic hydrolysis time 100 min,enzymatic hydrolysis temperature 55℃,liquid-to-material ratio 25∶1(mL/g)and enzymatic hydrolysis pH 5.0.Under the above conditions,the yield of polysaccharides was 5.18%,very close to the theoretical prediction from the regression model of 5.21%,the error was less than 5%.Polysaccharide from Curcuma zedoary had strong scavenging effects on DPPH radical,hydroxyl radical and superoxide anion radical,and their half inhibitory concentrations were 1.169 mg/mL,1.284 mg/mL and 1.076 mg/mL,respectively.Compound enzymatic method could significantly improve the yield of polysaccharide from Curcuma zedoary.The process is simple and feasible,and the polysaccharide obtained from Curcuma zedoary has antioxidant activity in vitro.
