摘 要:选取碱液提取和超声助碱提取两种方法,用正交实验结果进行对比,确定提取竹笋壳中不溶性膳食纤维的最佳提取工艺。以碱液提取和超声助碱提取两种方法单因素实验为基本依据,采用正交试验对比不同因素和两种不同提取方法对竹笋壳不溶性膳食纤维(IDF)得率和膨胀力的影响,并且采用红外光谱,扫描电镜、X-射线衍射表征方法对竹笋壳不溶性膳食纤维进行表征。结果表明,提取物与膳食纤维的结构相一致。竹笋壳的碱液提取条件为:碱液浓度1 mol/L,时间90 min,温度70℃,料液比1∶40(g/mL),膳食纤维提取率为55.67%,膨胀力为6.23 mL/g。超声助碱法提取条件:碱液浓度为1mol/L,超声功率80 W,超声时间为20 min,料液比为1∶15(g/mL),膳食纤维提取率为60.97%,膨胀力为6.26 mL/g。结论:用超声助碱法提取竹笋壳膳食纤维的得率比碱液提取法略高,本研究为竹笋壳提取膳食纤维相关开发研究提供一定的参考价值。Alkaline extraction and ultrasonic-assisted alkali extraction were compared;the best extraction process for extracting insoluble dietary fiber from bamboo shoot shells was determined by orthogonal experiments.Based on the single-factor experiments,orthogonal experiments were used to compare the effects of different factors on the yield and swelling force of bamboo shoot shell insoluble dietary fiber(IDF)from two different extraction methods.Infrared spectroscopy,scanning electron microscopy,X-ray diffraction were used to characterize the bamboo shoot shell insoluble dietary fiber.The results showed that the extract structure was consistent with that of dietary fiber.The optimal alkaline extraction conditions of bamboo shoot shells were:alkaline concentration 1 mol/L,extracting for 90 min.at 70℃,solid-liquid ratio 1∶40(g/mL).Under the above conditions,the dietary fiber extraction rate was 55.67%,and the dietary fiber expanding force was 6.23 mL/g.The optimal ultrasonic-assisted alkaline extraction conditions were:alkaline concentration 1 mol/L,ultrasonic power 80 W,ultrasonic time 20 min,solid-liquid ratio 1∶15(g/mL).The dietary fiber extraction rate under the above condition was 60.97%,and its expanding force was 6.26 mL/g.The yield of dietary fiber extracted from bamboo shoot shell by ultrasonic-assisted alkali method was slightly higher than that of alkaline extraction.This study provides a reference for the development and research on the extraction of dietary fi ber from bamboo shoot shell.
