基于膜分离技术的低聚乳果糖纯化及其对肠道菌群的影响

摘  要:选用截留分子质量3 ku的超滤膜组件和截留分子质量400 u的纳滤膜组件对粗低聚乳果糖(LS)进行分离纯化并进行工艺条件优化,将纯化后的LS与人体粪便共培养,探究其对肠道菌群的影响。结果显示,超滤工艺的最佳操作条件为:压力0.20 MPa,温度40℃,料液浓度10%;纳滤工艺的最佳操作条件为:压力0.55 MPa,温度40℃,料液稀释倍数为3倍左右。纯化后,LS含量显著提升至81.22%,产品终得率为86.72%。LS能增加肠道菌群的丰富度和多样性,并在属水平上显著提高双歧杆菌、栖粪杆菌等有益菌属的丰度,减少与肠性疾病相关的瘤胃球菌、志贺埃希氏菌等有害菌属的丰度,对肠道菌群具有良好的益生调节作用。The ultrafiltration membrane module with a molecular weight of 3 ku and the nanofiltration membrane module with a molecular weight of 400 u were selected to separate and purify crude lactofructose(LS)and the process conditions was optimized.The purified LS was co-cultured with human feces to explore its effect on intestinal microflora.The results showed that the optimal operating conditions of ultrafiltration process were as follows:operating pressure 0.20 MPa,operating temperature 40℃,feed concentration 10%;The optimal operating conditions of nanofiltration process were obtained:operating pressure 0.55 MPa,operating temperature 40℃,the dilution ratio of the feed solution about 3 times of the initial volume.After nanofiltration,the content of LS was significantly increased to 81.22%with the final yield of 86.72%.LS can increase the abundance and diversity of intestinal microflora,and significantly increase the abundance of harmful bacteria such as Bifidobacteria and Faecalibacterium at the genus level,and reduce the abundance of harmful bacteria such as Ruminococcus and Escherichia-Shigella associated with intestinal diseases,which show a good probiotic regulatory effect on intestinal flora.
