摘  要:粮食中二甲戊灵残留分析方法多为大型仪器检测,检测结果准确,但存在操作复杂、设备价格昂贵等问题,旨在建立一种基于免疫层析技术来检测粮食中二甲戊灵检测方法。通过研究,成功研制出一种选用时间分辨荧光微球为载体,利用二甲戊灵抗原抗体特异性反应来进行检测的快速定量检测卡,并摸索出二甲戊灵快速定量检测卡的最佳标记工艺为羊抗鼠抗体-145~198 nm微球-二甲戊灵抗体组合标记方案,最佳划膜液为PBS与CBS体系划膜液,最佳样品垫为GL0194与JL80,最佳样品稀释液体系为BBS体系。并对检测卡进行性能检测,得出其最低检出限为7.516μg/kg,最低定量限为19.873μg/kg,检测灵敏度高,可用于粮食中二甲戊灵的快速定量检测。Most of the analysis methods of Pendimethalin in grain are large-scale instrument detection methods.The detection results are accurate,but there are some problems such as complex operation and expensive equipment.This study aims to establish a detection method for Pendimethalin in grain based on immunochromatography.Through the research,a rapid quantitative detection card using time-resolved fluorescent microspheres as the carrier and the specific reaction of Pendimethalin antigen and antibody was successfully developed.It was found that the best marking process of Pendimethalin rapid quantitative detection card was combined Sheep anti mouse antibody and 145~198 nm microspheres and Pendimethalin antibody,the best cutting solution was PBS and CBS system,and the best binding pad was GL0194 and JL80,and the best sample dilution liquid is BBS system.The performance of the detection card is tested,and the minimum detection limit is 7.516μg/kg,the minimum quantitative limit is 19.873μg/kg,with high detection sensitivity,which can be used for Pendimethalin rapid quantitative detection in grain.
