摘  要:针对生物化学与分子生物学实验课程的特点,设计了一个将糖基转移酶和蔗糖合酶偶联制备红景天苷高的本科生物化学与分子生物学实验。首先,将糖基转移酶和蔗糖合酶分别在大肠杆菌中进行异源过量表达,然后将两种酶进行偶联,在糖基供体UDP-葡萄糖的存在下催化底物酪醇生成红景天苷。通过将该实验融入生物化学与分子生物学实验教学,不仅可以增强学生的科学素养和实践能力,还可以提高教学的质量和教学效果。In response to the characteristics of biochemistry and molecular biology laboratory courses,an undergraduate biochemistry and molecular biology experiment was designed to prepare salidroside by coupling the glycosyltransferase and sucrose synthase.First,the glycosyltransferase and sucrose synthase were separately overexpressed heterologously in Escherichia coli,and then the two enzymes were coupled to catalyze the substrate tyrosol to generate salidroside in the presence of the sugar donor UDP-glucose.By incorporating this experiment into biochemistry and molecular biology laboratory teaching,it can not only enhance students'scientific literacy and practical ability,but also improve the quality and effectiveness of teaching.
