摘  要:目的生物信息学分析YTH基因家族各成员在肾癌中的表达及功能。方法利用UALCAN、Kaplan-Meierplotter、cBioPortal、WebGestalt、GEPIA2、cBioPortal数据平台对YTH家族分子在肾癌患者中的表达差异、预后价值、基因改变、功能富集、免疫浸润相关性、甲基化进行生物信息学分析。结果肾癌中YTHDF2、YTHDF3和YTHDC1 mRNA表达均较正常肾组织明显下降,YTHDC2则较正常肾组织明显升高。YTHDF2和YTHDF3的低表达患者总生存期(OS)和无复发生存期(RFS)明显低于高表达患者。YTHDC2基因变异率为最高(占11%),主要遗传变异类型为基因扩增。富集分析显示YTH基因家族相关功能包括生物合成、分解及代谢。相关的信号通路有转化启动,mRNA代谢调控好喝基因表达转录调控等。YTH家族多数成员的表达与CD_(4)^(+)T,CD_(8)^(+)T细胞、内皮细胞的浸润呈显著性正相关,与巨噬细胞浸润呈负相关。YTHDF2、YTHDF3、YTHDC1在肾癌中的甲基化水平较正常肾组织中显著升高,YTHDC2甲基化水平则下降。结论YTH家族成员在肾癌中存在表达及功能差异。YTH相关基因特别是YTHDF2、YTHDF3分子可能成为肾癌诊断和预后评估的潜在的标志物。Objective explore the expression profiles and function of YTH family members based on bioinformatic approaches.Methods The expression profiles,prognostic value,function enrichment,gene alterations,association with immune cell infiltration,and methylation status of YTH family were analyzed by UALCAN,Kaplan-Meier plotter,cBioPortal,WebGestalt,GEPIA2,cBioPortal.Results The mRNA expression levels of YTHDC1,YTHDF2 and YTHDF3 in RCC were high while YTHDC2 was low than those in normal tissues.The patients with low expressions of YTHDF1 and YTHDF2 have shorter overall survival(OS)and relapse-free survival(RFS)than those high experession patients.The genetic alteration rate of YTHDC2 was the highest(11%).The main types of genetic variation is gene amplification.GO function enrichment analysis showed that the main functions of YTH family were biological regulation and metabolism process.Related signal pathways include translational initiation,mRNA metabolism regulation process,gene expression transcriptional regulation,etc.The expression of most members of the YTH family was positively correlated with the infiltration of CD_(4)^(+)T,CD_(8)^(+)T cells and endothelial cells,and negatively correlated with the infiltration of macrophages.The methylation status of YTHDF2,YTHDF3 and YTHDC1 in renal carcinoma was significantly higher than that in normal renal tissue,while the methylation status of YTHDC2 was decreased.Conclusion The expression profiles and function of YTH members were difference in RCC.YTH family members(especially YTHDF2 and YTHDF3)are may become the potential markers of renal cancer.
