摘 要:目的 探讨自然吸入细颗粒物(particulate matter 2.5,PM_(2.5))致小鼠肾脏损伤相关炎症因子及成纤维细胞生长因子21(fibroblast growth factor 21,FGF21)在肾脏中的表达情况,推断在自然吸入PM_(2.5)致肾脏损伤时FGF21是否有望成为潜在的生物学指标。方法 依据随机数字表法,将52只6周龄的健康雄性小鼠随机分为对照组(自然吸入空气)和实验组(PM_(2.5)染毒组),每组26只。实验组自然吸入PM_(2.5)气溶胶28周。HE染色观察小鼠肾脏病理形态,免疫组织化学法检测肾脏中肿瘤坏死因子α(Tumor necrosis factor-α,TNF-α)、白细胞介素1β(Interleukin 1β,IL-1β)、白细胞介素6(Interleukin 6,IL-6)及FGF21蛋白表达情况,并对FGF21与TNF-α、IL-1β和IL-6的相关性进行分析。结果 与对照组比较,实验组小鼠肾脏出现明显的病理损伤,差异有统计学意义(P<0.05);与对照组比较,实验组小鼠肾脏TNF-α、IL-1β和IL-6蛋白表达水平升高,FGF21蛋白表达水平下降,差异均有统计学意义(均P<0.05);FGF21与TNF-α、IL-1β和IL-6均存在负相关关系(均P<0.05)。结论 PM_(2.5)自然吸入可致小鼠肾脏发生明显损伤,抑制肾脏FGF21的表达,促进炎症因子TNF-α、IL-1β和IL-6的表达,FGF21与TNF-α、IL-1β和IL-6均存在负相关性。推测在自然吸入PM_(2.5)致肾脏损伤时FGF21有望成为潜在的生物学指标。Objective To investigate the expression of inflammatory factors and fibroblast growth factor 21(FGF21)in the kidneys of mice induced by fine natural inhalation of particulate matter _(2.5)(PM_(2.5)),inferwhether FGF21 is expected to be a potential biological indicator in kidney damage caused by natural inhalation of PM_(2.5).Methods According to the random number table method,52 healthy male mice aged 6 weeks were randomly divided into the control group(natural inhalation of air)and the experimental group(PM_(2.5) exposure group),with 26 mice in each group.The experimental group naturally inhaled PM2s aerosol for 28 weeks.HE staining was used to observe the pathological morphology of the mouse kidneys.The immunohistochemical method was used to detect the protein expressions of tumor necrosis factor-α(TNF-α),interleukin 1β(IL-1β),interleukin 6(IL-6)and fibroblast growth factor 21(FGF21).And the correlation between FCF21 and TNF-α,IL-1βand IL-6 was analyzed.Results Compared with the control group,the kidneys of the mice in the experimental group had obvious pathological damage,and the difference was statistically significant(P<0.05).Compared with the control group,the expression levels of TNF-α,IL-1βand IL-6 in the kidney of mice in the experimental group increased,while the expression level of FCF21 decreased,and the differences were statistically significant(all P<0.05).The FCF21 was negatively correlated with TNF-α,IL-1βand IL-6(all P<0.05).Conclusions The natural inhalation of PM2s can cause significant kidney damage in mice,inhibit the expression of FCF21 in the kidney,and promote the inflammatory factor TNF-α,IL-1βand IL-6 expression.The FCF21 is negatively correlated with TNF-α,IL-1βand IL-6.It is speculated that FGF21 may become a potential biological indicator in kidney damage caused by natural inhalation of PM_(2.5).
