摘 要:目的:从筛选太白贝母多糖的不同提取方法入手,比较不同生长年限的太白贝母多糖含量变化,并评价不同提取方法所得多糖的抗氧化活性。方法:采用苯酚-硫酸法,以葡萄糖为对照,比较热浸法、超声法、回流法三种提取方法得到的多糖含量,并在此基础上比较2~5年生太白贝母多糖含量;利用DPPH和ABTS自由基对不同提取方法的太白贝母多糖抗氧化活性进行比较。结果:在同等提取条件下,超声法得到太白贝母多糖含量最多(4.377%),热浸法次之(3.253%),煎煮法最少(2.893%);太白贝母在2~4年生长期内多糖含量随生长年限延长显著增加,4年生达最高(4.334%);超声法提取的多糖的抗氧化活性最强,且与V_(C)无明显差异,各提取方法下多糖的抗氧化性与浓度(0.2~1.2 mg/mL)呈正相关。结论:超声法提取的4年生太白贝母多糖的含量高且具有显著抗氧化活性。Objective:By screening different extraction methods of Fritillaria taipai-ensis P.Y.Li polysaccharide,the changes of polysaccharide content of F.taipaiensis at Different growth years were compared,and evaluated antioxidant activities of polysaccharides extracted by different extraction methods.Methods:Phenol-sulfuric acid method was used,with glucose as the control,to compare the polysaccharide content of three extraction methods,namely hot immersion,ultrasonic and reflux.Based on these methods,the content of polysaccharide in F.taipaiensis from 2 to 5 years old was compared.DPPH and ABTS free radicals were used to compare the antioxidant activities of polysaccharides from F.taipaiensis extracted by different methods.Results:Under the same extraction conditions,the ultrasonic method obtained the highest content of polysaccharides in F.taipaiensis(4.377%),followed by the hot dip method(3.253%),and the decoction method was the least(2.893%).The polysaccharide content of F.taipaien⁃sis increased significantly with the extension of growth years during the growth period of 2~4 years,and reached the highest in 4 years(4.334%).The antioxidant activity of polysaccharides extracted by ultrasonic method was the strongest,and there was no significant difference compared to V_(C).The antioxidant activity of polysaccharides obtained by each extraction method was positively correlated with the concentration(0.2~1.2 mg/mL).Conclusion:The polysaccharide content of 4‐year‐old F.taipaiensis was high and it had significant antioxidant activity.
