摘  要:为了优化菌丝培养基以期提高菌丝生物量和氨基酸产量,以荷叶离褶伞菌丝体为试验材料,以生物量和氨基酸含量为指标,利用Box-Behnken响应面法优化液体发酵培养基。试验结果表明:液体发酵最优培养基配方为马铃薯200 g/L、葡萄糖25 g/L、麸皮7.75 g/L、KH 2PO 41.5 g/L、MgSO 41.62 g/L;每100 mL液体培养基可得菌丝体生物量为1.096±0.025 g,菌球大小均匀、菌球浓密,且优化后的菌丝体总氨基酸含量提高了23.9%,子实体总氨基酸含量高达17.06%。优化培养基可用于荷叶离褶伞液体发酵工厂化生产。In order to optimize mycelium medium to improve mycelium biomass and amino acid yield,Box-Behnken response surface method was used to optimize liquid fermentation medium with mycelium from lotus leaf as test material and with biomass and amino acid content as indexes.The experimental results showed that the optimal medium formula for liquid fermentation included potato 200 g/L,glucose 25 g/L,bran 7.75 g/L,KH 2PO 41.5 g/L,and MgSO 41.62 g/L.The biomass of available mycelium per 100 mL of liquid medium was 1.096±0.025 g,the mycelium ball was uniform and dense,the total amino acid content of the optimized mycelium was increased by 23.9%,and the total amino acid content of the fruiting body was as high as 17.06%.The optimized medium can be used for the factory production of liquid strain of Lyophyllum decastes.
