摘  要:通过单因素试验和正交试验对超声波辅助纤维素酶法提取香菇柄中可溶性膳食纤维的提取工艺进行优化,并探究最优条件下提取的香菇柄可溶性膳食纤维(Soluble dietary fiber,SDF)和不溶性膳食纤维(Insoluble dietary fiber,IDF)理化性质及抗氧化活性。结果表明,香菇柄SDF最优提取条件为纤维素酶添加量1.0%、酶解pH7.0、酶解时间80 min,在此条件下,香菇柄SDF提取率为6.75%,SDF的持水力为5.16g/g,持油力为5.65g/g,膨胀性为6.6mL/g;IDF的持水力为5.63 g/g,持油力为6.92 g/g,膨胀性为2.8 mL/g;SDF的阳离子交换能力、葡萄糖吸附能力、抗氧化能力均优于IDF。超声波辅助纤维素酶法能显著提高香菇柄SDF得率(P<0.05),改善SDF的理化性质及抗氧化活性。The extraction process of soluble dietary fiber from Lentinus edodes stipe was optimized by ultrasonic-assisted cellulase method through single factor experiment and orthogonal experiment,then the physicochemical properties and antioxidant activity of soluble dietary fiber(SDF)and insoluble dietary fiber(IDF)under the optimal conditions were investigated.The results showed that the optimal extraction conditions of dietary fiber from Lentinus edodes stipe were as follows:cellulase dosage 1.0%,pH 7.0,enzymolysis time 80 min,and the extraction rate of SDF was 6.75%.The water holding capacity,oil holding capacity,and swelling capacity of SDF were 5.16 g/g,5.65 g/g and 6.6 mL/g,respectively.The water holding capacity,oil holding capacity and swelling capacity of IDF were 5.63 g/g,6.92 g/g and 2.8 mL/g,respectively.SDF had better cation exchange capacity,glucose adsorption capacity and antioxidant capacity than IDF.Ultrasonic-assisted cellulase method can significantly increase the yield of SDF from Lentinus edodes stipe(P<0.05),improve the physicochemical properties and antioxidant activity of SDF.
