摘 要:为研究广金钱草不同极性部位抗氧化及抗菌活性。采用石油醚、乙酸乙酯、水饱和正丁醇从广金钱草乙醇提取物萃取得到不同极性部位组分,并采用体外抗氧化活性和打孔法测定其抗氧化活性和抑菌活性。结果表明,醇提物中多酚含量为(7.074±0.312)mg/g,黄酮含量为(9.038±0.261)mg/g,石油醚部位、乙酸乙酯部位、正丁醇部位、水相萃取物中多酚含量依次为(11.167±0.175),(13.190±0.072),(12.776±0.037),(7.116±0.0378)mg GAE/g;黄酮含量范围为(9.038±0.261),(60.277±0.361),(80.697±0.121),(77.814±0.120),(24.823±0.150)mg CE/g。4种不同极性部位的DPPH自由基清除活性,铁离子还原力,ABTS自由基清除活性在(32.455±0.929)~(209.066±1.576)μmol TE/g,且均呈现出乙酸乙酯部位>正丁醇部位>石油醚部位>水部位的趋势。此外,广金钱草各部分部位物对大肠杆菌、金黄色葡萄球菌、沙门氏菌有较好的抗菌活性,也呈现出乙酸乙酯部位>正丁醇部位>石油醚部位>水相萃取部位的趋势。最小抑菌浓度结果表明,金黄色葡萄球菌和沙门氏菌对这4种极性溶剂萃取物的敏感性则较高,最不敏感的菌株是大肠杆菌。乙酸乙酯对3种细菌的MIC分别为6.25,25.00,6.25 mg/m L。广金钱草具有较好的抗氧化和抑菌活性,有望成为天然抗氧化和抑菌活性成分的主要来源。To study the antioxidant and antibacterial activity of different polar parts of Desmodium styracifolium(Osbeck)Merr.Petroleum ether,ethyl acetate and water-saturated n-butanol were used to extract the components of different polar parts from the ethanol extract of Desmodium styracifolium(Osbeck)Merr.,and the antioxidant activities and antibacterial activities were determined by in vitro and drilling method.The results showed that the polyphenols and flavonoids in the ethanol extract were(7.074±0.312)mg GAE/g and(9.038±0.261)mg CE/g,respectively.The polyphenol contents of petroleum ether fraction,ethyl acetate fraction,n-butanol fraction and aqueous phase extract were(11.167±0.175),(13.190±0.072),(12.776±0.037),(7.116±0.0378)mg GAE/g,respectively.The contents of flavonoids were(9.038±0.261),(60.277±0.361),(80.697±0.121),(77.814±0.120),(24.823±0.150)mg CE/g,respectively.DPPH radical scavenging activity,ferric reducing power and ABTS radical scavenging activity of four different polar fractions ranged from(32.455±0.929)to(209.066±1.576)μmol TE/g whit the trend of ethyl acetate fraction>n-butanol fraction>petroleum ether fraction>water fraction.In addition,all parts of Desmodium styracifolium(Osbeck)Merr.had good antibacterial activity against Escherichia coli,Staphylococcus aureus and Salmonella with the same trend of antioxidant activity.The results of minimum inhibitory concentration showed that Staphylococcus aureus and Salmonella were more sensitive to the four polar solvent extracts,and Escherichia coli was the least sensitive strain.The MIC of ethyl acetate against the three bacteria were 6.25,25.00,6.25 mg/mL,respectively.The plant had good antioxidant and antibacterial activities,and was expected to be the main source of natural antioxidant and antibacterial components.
