摘 要:目的分析头颈部鳞癌(含皮肤鳞癌)中基质金属蛋白酶1(MMP1)和微RNA(miRNA)表达水平并筛选目标miRNA。方法下载GEO相关数据,比较头颈部鳞癌MMP1表达水平。利用miRanda、TargetScan7.2和miRDB软件在线预测miRNA,进一步与miRTarBase结果做比对,获得目标miRNA并进行验证。分析目标miRNA与MMP1的相互关系。结果与正常组织或不典型增生病变相比,MMP1在头颈部鳞癌(含皮肤鳞癌)组织中高表达,MMP1可参与癌变整个发展过程,并促进HNSCC转移;在线预测共获得3个目标miRNAs:miR-202、miR-326和miR-488,其中miR-202 mirSVR评分最高。相反,与正常组织相比,miR-202头颈部鳞癌(含皮肤鳞癌)中,miR-202表达水平明显低于正常组织,从正常皮肤-KIN1/2-KIN3-cSCC,miR-202可参与疾病整个发展过程。相关分析显示miR-202表达与MMP1呈负相关(r=-0.14,P<0.05)。结论MMP1在头颈部鳞癌(含皮肤鳞癌)组织中高表达,miR-202低表达,miR-202可能是调节MMP1的真实miRNA,二者呈负相关。Objective To analyze the expression level of matrix metalloproteinase(MMP)1 and microRNA(miRNA)in head and neck squamous cell carcinoma(HNSCC)(including cutaneous squamous cell carcinoma,cSCC),and screen the target miRNA.Methods GEO datasets were downloaded and MMP1 expression level of HNSCC were further compared.Then miRanda,TargetScan7.2 and miRDB online software were employed to predict miRNAs,and to further compare with the miRTarBase results to obtain target miRNA and finally verify them.The correlation between the target miRNAs and MMP1 was also analyzed.Results Compared with normal tissue or atypical hyperplasia tissue,MMP1 was highly expressed in HNSCCs(including cSCCs).MMP1 participates in the whole process of tumorigenesis and promotes the metastasis of HNSCC.We obtained 3 targets miRNAs online including miR-202,miR-326 and miR-488,and the mirSVR score of miR-202 was the highest.On the contrary,the expression level of miR-202 in HNSCCs(including cSCCs)was significantly lower than that in normal tissues.From normal skin-KIN1/2-KIN3-cSCC,miR-202 participates in the whole development process of the disease.Correlation analysis showed that the expression of miR-202 was negatively correlated with MMP1(r=-0.14,P<0.05)Conclusion MMP1 is highly expressed in HNSCCs(including cSCCs).miR-202 may be the real miRNA regulating the expression of MMP1 negatively.
