摘  要:茶作为世界三大饮料之一,富含咖啡碱、类黄酮、茶氨酸等次生代谢产物,具有极高的营养价值。JAZ蛋白因含有TIFY保守序列而隶属于TIFY家族,是一类在植物发育和防御反应中起着关键作用的蛋白家族,也是茉莉酸信号途径中重要的负调控因子。为了探究JAZ基因在茶树中发挥的作用,本研究通过同源重组克隆技术从茶树中克隆到一个JAZ基因,命名为CsJAZ3。结合生物信息学、基因表达模式及相关性分析初步推测了该基因的生物学功能。本研究结果表明,CsJAZ3全长1 101 bp,编码366个氨基酸,属于亲水性蛋白质,不含信号肽,不属于分泌蛋白,无跨膜区域,定位在细胞核。该蛋白含有高度保守的结构域,具有典型的TIFY蛋白家族特征。CsJAZ3在茶树新梢各组织部位均有表达且存在显著差异,其中叶中表达量最高。进一步研究发现,CsJAZ3基因表达量与咖啡碱含量呈显著正相关,推测Cs JAZ3基因可能参与调控咖啡碱合成。本研究为进一步解析茶树JAZ基因功能及JA信号调控次生代谢产物合成机制提供参考。As one of the three major beverages in the world,tea is rich in secondary metabolites such as caffeine,flavonoids and theanine,which has extremely high nutritional value.JAZ protein belongs to TIFY family because of its conservative sequence,which plays a key role in plant development and defense response,and is also an im-portant negative regulatory factor in jasmonic acid signaling pathway.In order to explore the role of JAZ gene in tea plant,a JAZ gene named CsJAZ3 was cloned from tea plant by homologous recombination cloning technology.Combined with bioinformatics,gene expression pattern and correlation analysis,the biological function of the gene was preliminarily speculated.The results showed that CsJAZ3 was 1101 bp in length,encoding 366 amino acids.It belonged to hydrophilic protein,did not contain signal peptide,did not belong to secreted protein and had no transmembrane region,and was located in the nucleus.The protein contains highly conserved domains and has typical characteristics of TIFY protein family.CsJAZ3 was expressed in all tissues and parts of tea shoots,and there were significant differences,among which the expression in leaves was the highest.Further study found that the expression of CsJAZ3 gene was positively correlated with caffeine content,suggesting that CsJAZ3 gene might be involved in the regulation of caffeine synthesis.This study provides a reference for further understanding the function of JAZ gene in tea plant and the mechanism of JA signal regulating the synthesis of secondary metabo-lites.
