摘 要:试验旨在建立HPLC法测定莲黄颗粒中有效成分含量的方法,同时研究莲黄颗粒对沙门氏菌的体内外作用效果。采用e2695高效液相系统测定莲黄颗粒中4种有效成分;采用打孔法和鸡沙门氏菌感染模型法研究莲黄颗粒体内外对沙门氏菌的作用效果。结果显示,莲黄颗粒中没食子酸、秦皮甲素、黄芩苷、穿心莲内酯4种成分在测定的质量浓度范围内线性关系良好,R^(2)均大于0.999,回收率在97%~103%之间,RSD值均小于2.5%。建立的HPLC检测方法简单、高效、灵敏度好。莲黄颗粒体外对沙门氏菌的抑制为高敏,体内能改善沙门氏菌感染导致鸡采食量的降低,并能修复肝脏和盲肠的损伤。研究为莲黄颗粒质量评价和临床应用提供数据支持。The study was to establish a HPLC method for the determination of the content of active ingredients in Lianhuang granules,and to study the effect of Lianhuang granules on Salmonella in vitro and in vivo.The e2695 high performance liquid phase system was used to determine four active components in Lianhuang granules.The effect of Lianhuang granules on Salmonella in vitro and in vivo was studied by the method of perforation and the model ofSalmonella infection in chickens.The results showed that the linear relationship of gallic acid,aesculin,baicalin and and rographolide in Lianhuang granules was good within the determined mass concentration range.The R^(2) was greater than 0.999,the recovery rate was between 97%and 103%,and the RSD value was less than 2.5%.The established HPLC method was simple,efficient and sensitive.Lianhuang granule was highly sensitive to Salmonella in vitro,and can improve the reduction of feed intake caused by Salmonella infection in vivo and repair the damage of Salmonella to liver and cecum.The study provides data support for the quality evaluation and clinical application of Lianhuang granule.
