摘 要:SPAC27E2.11c基因编码的是一种尚未被正式命名的粟酒裂殖酵母(Schizosaccharomyces pombe)特异性蛋白,该蛋白定位于线粒体质膜外侧.采用活细胞成像技术,研究粟酒裂殖酵母SPAC27E2.11c基因缺失后有丝分裂不同时期微管动力学的变化情况.结果表明:SPAC27E2.11c基因缺失导致间期含5根微管束的细胞比例增加,含3根微管束的细胞比例减少,并出现含2根微管束的细胞;间期微管束长度增加,收缩速率降低,且微管束在细胞壁停留时间延长;前期“棒状”纺锤体形成减少并出现单极纺锤体;中期纺锤体伸长速率降低,伸长时间延长,提示纺锤体组装检查点被激活;后期纺锤体“拱型”断裂减少,“直线型”断裂增加,“S型”断裂消失.SPAC27E2.11c基因缺失还导致染色体不完全分离向细胞两极及染色体分离后细胞不分裂的异常情况.以上结果表明SPAC27E2.11c基因缺失导致间期微管动力学失衡,分裂期纺锤体形成和断裂缺陷及染色体分离缺陷.SPAC27E2.11c gene encodes a specific protein of Schizosaccharomyces pombe that is located in the mitochondrial outer membrane and has not been officially named.In this study,the experimental technology of living cell imaging was applied to study the effect of S.pombe SPAC27E2.11c gene deletion on microtubule dynamics during different stages of mitosis.The results showed that SPAC27E2.11c gene deletion increased the proportion of cells containing 5 microtubule bundles,decreased the proportion of cells containing 3 microtubule bundles,and produced cells containing 2 microtubule bundles.During the interphase,the length of microtubules increased,the contraction rate decreased and the residence time of microtubule bundles in cell wall prolonged.During the prophase,the formation of"rod"spindle decreased and unipolar spindle formed.During the metaphase,the spindle elongation rate decreased and the elongation time prolonged,indicating the activation of spindle assembly checkpoint.During the anaphase,the"arch type"fracture of spindle decreased,the"linear type"fracture increased,and the"S-type"fracture disappeared.The deletion of SPAC27E2.11c gene also led to the abnormal situation of incomplete chromosome separation to the two poles of cells and non division of cells after chromosome separation.The above results suggested that SPAC27E2.11c gene deletion led to the imbalance of microtubule dynamics in interphase,the defects of spindle formation and breakage and chromosome segregation in mitosis.
