摘  要:目的探讨电针对佐剂性关节炎模型大鼠脾脏CD4^(+)T淋巴细胞增殖的抑制作用及其对G6PD/NADPH信号通路的影响。方法将27只SD雄性大鼠随机分为3组(n=9):空白组、模型组、电针组。模型组、电针组造模向大鼠尾根部皮下注射不完全弗氏佐剂建立AA大鼠模型。电针组施以电针“足三里”、“关元”、“阿是穴”,治疗每天1次,时间30min,持续3周。利用足跖容积测量仪测定大鼠足趾肿胀度;运用苏木精-伊红染色观察大鼠滑膜组织细胞形态学变化;采用流式细胞术检测脾脏中CD4^(+)T细胞数量;使用Western blot法和ELISA法检测大鼠脾脏和血浆中G6PD和NADPH的表达量。结果相较于空白组,模型组脾脏中CD4^(+)T细胞数量,血浆和脾脏中G6PD、NADPH的表达量显著升高(P<0.01);相较于模型组,电针组脾脏中CD4^(+)T细胞数量,血浆和脾脏中G6PD、NADPH的表达量显著降低(P<0.01或P<0.05)。结论电针“足三里”、“关元”、“阿是穴”可减少AA大鼠关节滑膜炎症的产生,抑制CD4^(+)T增殖,其机制可能与电针抑制磷酸戊糖途径关键酶G6PD和其产物的NADPH的表达相关。Objective To investigate the inhibitory effect of electroacupuncture on the proliferation of CD4^(+)T lymphocytes in spleen of rats with adjuvant arthritis and its effect on the G6PD/NADPH signaling pathway.Methods Twenty-seven male SD rats were randomly divided into three groups(n=9),blank group,model group and electroacupuncture group.The rat model of AA was established by subcutaneous injection of incomplete Freund's incomplete adjuvant into the tail root.Electroacupuncture group was given acupuncture at“Zusanli”(ST36),“Guanyuan”(RN4)and“A’shi”(experience acupoint)on both sides once a day for 30 minutes for 3 weeks.Toe swelling of rats was measured by plantar volume measuring instrument.The morphological changes of rat synovial tissue cells were observed by hematoxylin-eosin staining.The number of CD4^(+)T cells in spleen was measured by flow cytometry.Western blot and ELISA were applied to detect the content of G6PD and NADPH in spleen and plas-ma of rats.Results Compared with blank group,the number of CD4^(+)T cells in spleen,the content of G6PD and NADPH in plasma and spleen were significantly increased in model group(P<0.01).Compared with model group,the number of CD4^(+)T cells in spleen and the content of G6PD and NADPH in plasma and spleen were significantly decreased in electroacupuncture group(P<0.01 or P<0.05).Conclusion Electroacupuncture at"Zusanli","Guyuan"and"A’shi-point"can reduce syno-vial inflammation and inhibit the proliferation of CD4^(+)T cells in AA rats.The mechanism may be related to the inhibition of the expression of G6PD,a key enzyme in pentose phosphate pathway,and NADPH by electroacupuncture.
