摘 要:目的观察骨软骨瘤组织、骨肉瘤组织中Fascin和沉默信息调节因子2(SIRT2)的表达水平, 探讨Fascin和SIRT2表达与骨肉瘤患者临床特征的关系。方法收集2019年1月至2021年12月山西省人民医院保存的82例骨肉瘤组织和39例骨软骨瘤组织标本作为本项目研究对象, 采用免疫组织化学法检测上述组织中Fascin和SIRT2的表达, 结合临床资料, 采用χ2检验分析Fascin和SIRT2表达水平与骨肉瘤患者临床病理参数的相关性。结果骨肉瘤组织中Fascin阳性表达率为74.39%(61/82), 明显高于骨软骨瘤组织中Fascin阳性表达率[23.08%(9/39)], 差异有统计学意义(χ2=28.540, P<0.01)。骨肉瘤组织中SIRT2阳性表达率为65.85%(54/82), 明显高于骨软骨瘤组织中SIRT2阳性表达率[15.39%(6/39)], 差异有统计学意义(χ2=26.930, P<0.01)。Fascin表达水平与骨肉瘤患者软组织浸润、Eneeking分期、肺转移明显相关(χ2=4.952、5.844、6.385, P<0.05), Fascin表达水平与骨肉瘤患者病变部位、性别、组织类型、碱性磷酸酶(ALP)水平、年龄无相关(χ2=0.002、0.080、0.059、0.035、1.590, P>0.05)。SIRT2表达水平与骨肉瘤患者Eneeking分期、肺转移明显相关(χ2=5.017、4.748, P<0.05), SIRT2表达水平与骨肉瘤患者病变部位、性别、软组织浸润、组织类型、年龄、ALP水平无相关(χ2=0.012、0.016、0.016、0.004、0.196、0.548, P>0.05)。结论骨肉瘤组织中Fascin和SIRT2表达上调, 对骨肉瘤患者进行Fascin和SIRT2的检测可评估骨肉瘤患者肿瘤的恶性程度并判断患者预后。Objective To investigate the expression levels of Fascin and silentmating information regulater 2(SIRT2)in osteochondroma and osteosarcoma,and to explore the relationship between Fascin and SIRT2 expression and the clinical characteristics of osteosarcoma patients.Methods A total of 82 cases of osteosarcoma tissue and 39 cases of osteochondroma tissue specimens preserved in Shanxi Provincial People’s Hospital from January 2019 to December 2021 were collected as the research objects.The expression of Fascin and SIRT2 in the above tissues was detected by immunohistochemistry.Chi-square test was used to analyze the correlation between Fascin and SIRT2 expression levels and clinicopathological parameters of osteosarcoma patients.Results The positive expression rate of Fascin in osteosarcoma tissues was 74.39%(61/82),which was significantly higher than that in osteochondroma tissues[23.08%(9/39)](χ2=28.540,P<0.01).The positive expression rate of SIRT2 in osteosarcoma tissues was 65.85%(54/82),which was significantly higher than that in osteochondroma tissues[15.39%(6/39)](χ2=26.930,P<0.01).Fascin expression level was significantly correlated with soft tissue invasion,Eneeking stage and lung metastasis in osteosarcoma patients(respectively,χ2=4.952,5.844,6.385,P<0.05),but not significantly with lesion location,gender,tissue type,alkaline phosphatase(ALP)level and age in osteosarcoma patients(respectively,χ2=0.002,0.080,0.059,0.035,1.590,P>0.05).The expression level of SIRT2 was significantly correlated with Eneeking stage and lung metastasis of osteosarcoma patients(respectively,χ2=5.017,4.748,P<0.05),but not significantly with lesion location,gender,soft tissue infiltration,tissue type,age and ALP level in patients with osteosarcoma(respectively,χ2=0.012,0.016,0.016,0.004,0.196,0.548,P>0.05).Conclusion The expression levels of Fascin and SIRT2 are up-regulated in osteosarcoma tissues.The detection of Fascin and SIRT2 in patients with osteosarcoma can evaluate the malignant degree of osteosarcoma and determine
