接骨木镰刀菌产呕吐毒素条件筛选及致病性分析

摘  要:旨在通过单因素试验对接骨木镰刀菌产呕吐毒素的温度、时间、pH、碳源及碳源浓度进行筛选,应用响应面法设计试验,依据Box-Behnken中心组合原理筛选得到影响接骨木镰刀菌产呕吐毒素的关键因素。以呕吐毒素含量为响应值借助Design-Expert v8.0.6软件分析各影响因子与响应值的相互作用,预测最适条件并验证。试验得到一个拟合程度高、误差小的模型,得出呕吐毒素含量最大时的条件为:葡萄糖为碳源、培养温度26.19℃、pH 6.12、培养时间25.38 d。在此条件下,呕吐毒素含量预测值为2.50 ng/mL,实际测得呕吐毒素含量为2.57 ng/mL。低浓度呕吐毒素处理能减小马铃薯病斑面积20%。In this study,we explored the impact of various factors on production of vomitoxin by Fusarium sambucinum,including temperature,time,pH,carbon sourceand carbon source concentration.To design the experiment,we employed response surface methodology,and screened the key factors affecting the production of vomitoxin by Fusarium sambucinum using the Box-Behnken center combination principle.The response value of vomitoxin content was analyzed using Design-Expert v8.0.6 software to determine the optimum conditions and verify it.By experimenting with the interaction of each influencing factor on the response value,a model with high fit and low error was obtained.The conditions for obtaining maximal vomitoxin content was determined to be glucose as carbon source,an incubation temperature of 26.19℃,pH 6.12,and an incubation time of 25.38 d.Under these conditions,the predicted vomitoxin content was 2.50 ng/mL,and the actual vomitoxin content was 2.57 ng/ml,treatment with low concentrations of vomitoxin reduced potato lesion area by 20%.
