摘 要:目的探讨阿魏酸(FA)在烟曲霉菌诱导的人角膜上皮细胞中的抗炎作用。方法采用CCK-8方法检测不同浓度FA(0、50、100、150、200、250、300μmol/L)溶液对人角膜上皮细胞(HCEC)活力的影响;采用RT-PCR方法检测灭活的烟曲霉菌菌丝感染HCEC后,FA对炎症因子白细胞介素1β(IL-1β)、白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)及核因子E2相关因子2(Nrf2)mRNA表达的影响;采用ELISA方法检测FA对炎症因子IL-6及单核细胞趋化蛋白(MCP-1)蛋白表达的影响。结果300μmol/L的FA溶液处理组HCEC存活率低于其他各组(F=5.390,P<0.05),250μmol/L及以下浓度FA溶液处理对HCEC活力无影响(P>0.05)。250μmol/L FA处理可显著升高Nrf2 mRNA表达,降低IL-1β、IL-6、TNF-αmRNA的表达水平,并显著降低IL-6及MCP-1蛋白表达(F=4.720~990.967,P<0.05)。结论FA可能通过上调Nrf2的表达,抑制烟曲霉菌诱导的HCEC炎症反应中促炎因子的表达,发挥抗炎作用。Objective Toinvestigatethe anti-inflammatory effect of ferulic acid(FA)in human corneal epithelial cells induced by Aspergillus fumigatus.Methods CCK-8 assay was used to observe the effect of different concentrations of FA solution(0,50,100,150,200,250,300μmol/L)on the viability of human corneal epithelial cells(HCEC);RT-PCR was used to measure the effect of FA on the mRNA expression levels of the inflammatory factors interleukin-1β(IL-1β),interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),and nuclear factor erythroid 2-related factor 2(Nrf2)after HCEC were infected with inactivated Aspergillus fumigatus hyphae;ELISA was used to observethe effect of FA on the protein expression levels of the inflammatory factors IL-6 and monocyte chemotactic protein-1(MCP-1).Results The group of HCEC cells treated by 300μmol/L FA solution had a significantly lower cell viabilitythan the other groups(F=5.390,P<0.05),and FA solution with a concentration of 250μmol/L or less had no effect on the viability of HCEC(P>0.05).Treatment with 250μmol/L FA significantly increased the mRNA expression levelof Nrf2 and significantly reduced the mRNA expression levels of IL-1β,IL-6,TNF-α,and the protein expression level of MCP-1(F=4.720-990.967,P<0.05).Conclusion FA may exert an anti-inflammatory effect by upregulating the expression of Nrf2 and inhibiting the expression of pro-inflammatory factors in the inflammatory response of HCEC induced by Aspergillus fumigatus.
