富硒绞股蓝的抗氧化活性研究

摘  要:采用酸性氧化电解水预处理-乙醇回流提取两步法提取富硒绞股蓝与普通绞股蓝活性物质,用荧光光度法测定两者提取物的硒元素含量分别为0.21μg/g和0.06μg/g。以DPPH、羟基和超氧阴离子自由基清除率为指标,比较不同浓度富硒与普通绞股蓝提取物的抗氧化活性,发现前者对自由基的清除率皆高于后者。在提取液浓度最高(1.0 mg/mL)时,富硒绞股蓝对3种自由基的清除率比普通绞股蓝分别提高了1.3%、6.07%和11.73%。通过在普通绞股蓝提取液中添加不同浓度硒代蛋氨酸,验证了硒元素是富硒绞股蓝比普通绞股蓝具有更显著抗氧化效果的原因。The active substances of selenium-rich Gynostemma pentaphyllum and common Gynostemma pentaphyllum were extracted by the two-step method of acid oxidation electrolysed water pretreatment and ethanol reflux extraction.The selenium content of the two extracts was determined to be 0.21μg/g and 0.06μg/g respectively by fluorescence spectrophotometry.Taking DPPH,hydroxyl and superoxide anion radical scavenging rates as indicators,the antioxidant activities of selenium-rich Gynostemma pentaphyllum extract and common Gynostemma pentaphyllum extract at different concentrations were compared.It was found that the scavenging rates of free radicals of the former were higher than those of the latter.When the concentration of extract was the highest(1.0 mg/mL),the scavenging rate of seleniumrich Gynostemma pentaphyllum on three free radicals was 1.3%,6.07% and 11.73% higher than that of common Gynostemma pentaphyllum respectively.By adding different concentrations of selenomethionine in the extract of common Gynostemma pentaphyllum,it was verified that selenium was the reason why selenium-rich Gynostemma pentaphyllum had more significant antioxidant effect than common Gynostemma pentaphyllum.
