内生菌对镉胁迫蚕豆根尖细胞毒理效应的影响

摘  要:镉(Cd)作为造成农业污染的重金属之一,对大多数植物的生长有很大影响。越来越多的研究表明植物内生菌与植物互作能够显著提高宿主植物对重金属胁迫的耐性。先用野大豆内生菌YD25对蚕豆根尖进行侵染,将其分为侵染组(E+)和未侵染组(E-),再分别同时用0,4和8 mg·L^(-1)的CdCl_(2)溶液进行镉胁迫处理,然后采用蚕豆根尖微核技术,对蚕豆根尖细胞有丝分裂指数、染色体畸变率及微核率进行测定。结果表明Cd^(2+)对蚕豆根尖细胞有毒害作用。随着溶液浓度的增加,有丝分裂指数下降,染色体畸变率和微核率上升,抑制效应逐渐增强。在所有处理中,E+组的有丝分裂指数高于E-组,E+组的染色体畸变率及微核率均低于E-组,说明内生菌YD25的侵染缓解了镉对蚕豆根尖细胞的毒害效应。Cadmium(Cd),as one of the heavy metals causing agricultural pollution,has a great effect on the growth of most plants.More and more studies have shown that the interaction of endophytic fungi with plants can significantly improve the tolerance of host plants to heavy metal stress.In this study,the endophytic fungus YD25 of wild soybean was used to dye the root tips of broad bean,which were divided into the infected group(E+)and the uninfected group(E-).At the same time,CdCl_(2)solutions of 0 mg·L^(-1),4 mg·L^(-1)and 8 mg·L^(-1)were used to treat cadmium stress,respectively.Micromodular methods were used to determine mitotic index,chromosome aberration and root cell micromodules.The results show that Cd^(2+)is toxic to viciafaba root tip cells.With the increase of solution concentration,the mitotic index decreases,the chromosome aberration rate and micronucleus rate increases,and the inhibitory effect is gradually enhanced.In all treatments,the mitotic index of E+group is higher than that of E-group.The chromosomal aberration rate and micronucleus rate of E+group are lower than those of E-group,indicating that the exposure of endophytic fungus YD25 alleviates the toxic effect of cadmium on viciafaba root tip cells.
