摘  要:从糙皮侧耳基因组中预测获得PoEgt2基因进行生物信息学分析和表达分析。结果显示:PoEgt2编码氨基酸546个,为胞内不稳定蛋白,无跨膜区和信号肽。蛋白质序列比对结果显示,糙皮侧耳PoEgt2与白黄侧耳、刺芹侧耳等侧耳属真菌同源性较高,与非侧耳属菌同源性较低。PoEgt2在糙皮侧耳发酵前期的表达量较高,于发酵4 d达到峰值,随后表达水平随时间下降。该研究可为高产麦角硫因的糙皮侧耳菌种改良和发酵条件优化提供科学依据。In this study,PoEgt2 was predicted from Pleurotus ostreatus genome for bioinformatics analysis and expression analysis.The results showed that PoEgt2 encoded 546 amino acids,which was an intracellular unstable protein without transmembrane region and signal peptide.The results of protein sequence alignment showed that PoEgt2 had high homology with P.cornucopiae and P.eryngii,but low homology with non-Pleuloplegia.The expression level of PoEgt2 was higher in the early stage of fermentation and reached its peak on the fourth day of fermentation,and then decreased with time.This study provided a scientific basis for the improvement of Pleurotus ostreatus strains with high ergothioneine yield and optimization of fermentation conditions.
