摘  要:[目的]研究超声辅助提取桂林产黑老虎果皮总黄酮的最佳工艺以及提取物对DPPH、ABTS+自由基的清除能力。[方法]通过单因素试验(乙醇体积分数、料液比、超声时间、超声温度)及正交试验优化总黄酮的提取工艺,并测定提取物对DPPH、ABTS+自由基的清除能力。[结果]桂林产黑老虎果皮总黄酮的最佳工艺条件为40%乙醇、料液比1∶40(g∶mL)、超声提取60 min、超声温度60℃,该条件下平行提取6次得到总黄酮的提取率为(22.48±0.40)mg/g,RSD为1.78%。桂林产黑老虎果皮总黄酮对DPPH、ABTS+自由基均有一定的清除能力,IC_(50)分别为204.57和116.03μg/mL,AEAC分别为43.21和129.95 mg/g。[结论]该提取方法可行、重复性好。黑老虎果皮总黄酮在体外具有良好的DPPH、ABTS+自由基清除能力。[Objective]To study the optimal extraction conditions of ultrasonic-assisted extraction and the scavenging effects on DPPH and ABTS+of total flavonoids from Kadsura coccinea pericarp of Guilin.[Method]The extraction process of total flavonoids was optimized by single factor experiments(ethanol volume fraction,solid-liquid ratio,ultrasound time,ultrasound temperature)and orthogonal experiments,and the scavenging ability of the extract on DPPH,ABTS+free radicals were determined.[Result]The optimal process conditions for producing total flavonoids from Kadsura coccinea pericarp in Guilin were 40%ethanol,a material liquid ratio of 1∶40(g∶mL),ultrasonic extraction for 60 minutes,and ultrasonic temperature of 60℃.Under this condition,the extraction rate of total flavonoids obtained by parallel extraction for 6 times was(22.48±0.40)mg/g,and RSD was 1.78%(n=6).The total flavonoids had certain scavenging capacity for DPPH and ABTS+radicals,with IC_(50) of 204.57 and 116.03μg/mL,and AEAC of 43.12 and 129.95 mg/g,respectively.[Conclusion]The extraction method is feasible and has good reproducibility.Total flavonoids from Kadsura coccinea pericarp of Guilin has good DPPH and ABTS+free radical scavenging ability in vitro.
