摘 要:以荷叶为原料,在单因素实验基础上通过响应面法优化,得到超声辅助离子液体提取荷叶黄酮的工艺参数,并评价其抗氧化活性。结果表明,荷叶黄酮的最佳提取工艺为:0.9 mol/L的1-己基-3-甲基咪唑四氟硼酸盐([C6mim]BF4)水溶液作为提取溶剂,固液比1∶25(g∶m L),超声功率186 W,超声时间24 min,提取温度70℃。在该条件下,荷叶黄酮提取率为4.65%。荷叶黄酮对1,1-二苯基-2-三硝基苯肼自由基(DPPH·)、·OH具有良好的清除能力,半数抑制浓度(IC50)分别为0.8847和1.1445 g/L。荷叶黄酮具有较强的还原力,当质量浓度为1.6 g/L时,吸光度最大值为0.72 a.u.;荷叶黄酮对猪油和花生油的氧化均有一定的抑制作用,表现出良好的抗氧化活性。Ionic liquid-based ultrasonic assisted extraction parameters of flavonoids from lotus leaf were optimized via single factor experiment and response surface method.The flavonoids obtained was then evaluated for its antioxidant activity.The results showed the optimal conditions were as follows:0.9 mol/L 1-hexyl-3-methylimidazole tetrafluoroborate([C6mim]BF4)aqueous solution as extraction solvent,solid-liquid ratio of 1∶25(g∶mL),ultrasonic power of 186 W,ultrasonic time of 24 min,and extraction temperature of 70℃.The extraction rate of lotus leaf flavonoids reached 4.65%under the optimized conditions.The extracted lotus leaf flavonoids showed good antioxidant activity for 1,1-diphenyl-2-trinitrophenylhydrazine free radical(DPPH•)and•OH,with a corresponding half inhibitory concentration(IC50)of 0.8847 and 1.1445 g/L respectively.The lotus leaf flavonoids displayed strong reducing power with the maximum absorbance of 0.72 a.u.at a mass concentration of 1.6 g/L.The lotus leaf flavonoids exhibited good inhibitory effect on the oxidation of lard and peanut oil,showing good antioxidant activity.
