摘 要:以水提醇沉法提取紫苏多糖(Perilla frutescens polysaccharides,PFPS),经初级分离后,采用HNO_(3)-Na_(2)SeO_(3)法对PFPS进行硒酸改性修饰得到硒化-紫苏多糖(Se-PFPS),并采用傅里叶变换红外光谱(Fourier transform infrared spectroscopy,FTIR)、纳米粒度仪、高效凝胶渗透色谱(high performance gel permeation chromatography,HPGPC)、扫描电子显微镜(scanning electron microscopy,SEM)、X射线衍射(X-ray diffraction,XRD)仪和热重分析仪(thermal gravimetric analyzer,TGA)对2种多糖进行了理化及结构特性分析。原子荧光光谱分析显示硒化-紫苏多糖(Se-PFPS)硒含量可达(1004.33±48.60)μg/g;FTIR分析显示Se-PFPS在620 cm^(-1)处存在Se—O—C特殊吸收峰,表明Na2SeO3与PFPS发生了化学结合,且以亚硒酸酯的形式存在;HPGPC、粒径分析显示化学修饰减小了多糖的颗粒尺寸,但同时增加了多糖分子质量;SEM和XRD分析显示硒酸改性改变了多糖的形态特征和晶体状态,且多以多晶和非晶形式存在;TGA分析显示PFPS比Se-PFPS的热稳定性更强。In this study,polysaccharides(PFPS)were extracted form Perilla frutescens L.and modified by HNO_(3)-Na_(2)SeO_(3) method,and the physicochemical and structural characteristics of PFPS and selenized PFPS(Se-PFPS)was analyzed by Fourier transform infrared(FTIR)spectroscopy,nanoparticle size analyzer,high performance gel permeation chromatography(HPGPC),scanning electron microscopy(SEM),X-ray diffraction(XRD)and thermal gravimetric analyzer(TGA).The selenium content of Se-PFPS was(1004.33±48.60)μg/g as determined by atomic fluorescence spectrometry(AFS).The FTIR spectrum showed a characteristic absorption peak of Se‒O‒C at 620 cm^(-1),indicating that sodium selenite was chemically bound to PFPS and seleniumin in Se-PFPS existed in the form of selenite.HPGPC and particle size analysis showed that selenization modification reduced the particle size,and increased the molecular mass of PFPS.The results of SEM and XRD showed that selenization modification changed the morphology and crystalline state of PFPS,and Se-PFPS mainly existed in polycrystalline and amorphous forms.TGA analysis showed that PFPS had stronger thermal stability than did Se-PFPS.
