摘  要:透明质酸(Hyaluronic acid, HA)是一种聚电解质,很难溶解于非极性溶剂,溶解于水溶液时由于电荷排斥作用,导致溶解过程中液体黏度大,较长时间才能完成溶解。为了促进HA的快速溶解,制备了泡腾颗粒并确定其最佳制备工艺。首先,对泡腾颗粒辅料进行选择确定,其次,通过单因素试验筛选湿法制粒所使用的润湿剂浓度、柠檬酸与碳酸氢钠的质量分数、柠檬酸与碳酸氢钠的比例、甜味剂添加量、香精添加量,最后通过验证试验确认工艺的重现性。最佳工艺:以柠檬酸22%、碳酸氢钠20%、木糖醇18%、异麦芽酮糖醇20%、甜菊糖苷1.8 g/kg、香精1.4g/100 g制备泡腾颗粒。该工艺简单,且能实现高剂量透明质酸在产品体系中的快速溶解。Hyaluronic acid(HA)is a kind of polyelectrolyte,which is dificult to dissolve in non-polar solvents.In aqueous solution,due to charge repulsion,a great volume repulsion effect is formed,and the solution formed by dissolution has a great viscosity,which takes a long time to dissolve completely.If HA is simply mixed with other excipients,it is easy to form a group in the process of adding water to dissolve,and the dissolution time is longer.In order to promote the rapid dissolution of HA,effervescent granules are prepared,and the best preparation process is determined.Firstly,the ingredients of effervescent granules are selected and determined.Secondly,the concentration of adhesive used in wet granulation,the mass fraction of citric acid and sodium bicarbonate,the ratio of citric acid to sodium bicarbonate,the amount of sweetener added and the amount of essence added are screened by single factor experiment.Finally,the reproducibility of the process is confirmed by verification experiments.The optimal process is to prepare effervescent granules with 22%citric acid,20%sodium bicarbonate,18%xylitol,20%isomalt,1.8 g/kg stevia glycoside and 1.4 g/100 g essence.The process is simple and can realize the rapid dissolution of high dose hyaluronic acid in the product system.
