摘  要:采用高效液相色谱法(HPLC)测定腐乳中红曲红胺、红曲素和红曲红素含量。用无水乙醇直接提取,Silversil C18色谱柱在流速1.0 mL/min,40℃条件下,通过流动相[甲醇和乙酸-乙酸铵溶液(pH 5.0)]进行梯度洗脱。检测波长分别为:红曲红胺264 nm;红曲素和红曲红素为390 nm。进样量20μL。3种红曲色素相关系数R^(2)为0.99990~0.99998,线性关系良好。检出限为0.0312~28.8 mg/kg,定量限为0.0104~96.0 mg/kg。加标质量分数在1~5000 mg/kg时,方法回收率为91.5%~101.8%,相对标准偏差(n=6)为0.36%~1.36%。该方法精密度良好,准确度高,可适用于腐乳中红曲红胺、红曲素和红曲红素含量的测定。Monascus amine,monascin and monascus red in preserved beancurd by a high performance liquid chromatography(HPLC)method were determined.Samples were extracted with anhydrous ethyl alcohol on Silversil Cis column.The flow rate was 1.0 mL/min,and the separation was carried out at 40℃.Methanol and acetic acid-ammonium acetate solution(pH 5.0)were used as mobile phase with gradient elution.The detection wavelength of monascus amine was 264 nm.The detection wavelength of monascin and monascus red was 264 nm.The injection volume was 20μL.The correlation coeficient R^(2) of three monascus pigments in the concentration range was 0.99990-0.99998,indicating a good linear relationship.The limits of detection were 0.0312-28.8 mg/kg and the limits of quantification were 0.0104-96.0 mg/kg.The recoveries were 91.5%-101.8%when the standards were 1-5000 mg/kg,and the relative standard deviations(n=6)were 0.36%-1.36%.The method had good precision and accuracy,which can be used for the determination of monascus amine,monascin and monascus red in preserved beancurd.
