摘 要:以罗非鱼鱼皮明胶为原料,经胰蛋白酶的酶解作用得到胶原蛋白肽,对其进行酶解工艺优化。以肽得率作为指标,采用单因素试验得出明胶浓度、酶用量、温度和pH值随时间的延长对肽得率的影响;在单因素试验的基础上,采用Central-composite法对其酶解工艺进行优化,以酶用量、缓冲溶液pH值、明胶浓度、温度和时间为自变量,以明胶酶解后肽得率为响应面值进行对明胶酶解工艺优化分析。确定最佳酶解工艺条件为明胶底物添加量6.0%,酶用量15.1μg/g,酶解温度70.00℃,pH值7.63和时间69.99 min,在此最佳工艺条件下酶解罗非鱼皮明胶肽得率76.54%;最后根据结果分析与实际结合,把明胶酶解优化条件定为酶解温度70.0℃,明胶添加量6.0%,反应pH值8.00,时间70.0 min,酶用量为20.0μg/g时,明胶酶解的实际肽得率为77.72%,与理论预测值的误差约为1.18%。由此可见,建立的响应面模型是可靠的,具有实用参考价值,可在实际生产中推广使用。Taking the gelatin of tilapia skin as raw material,collagen peptide was obtained by enzymatic hydrolysis of trypsin,and its enzymolysis process was optimized.Using the peptide yield as an indicator,single factor experiments were used to determine the effect of gelatin concentration,enzyme dosage,temperature,and pH over time on peptide yield.On the basis of single factor experiments,the enzymatic hydrolysis process was optimized using the Central-Composite method.The enzyme dosage,buffer solution pH,gelatin concentration,temperature and time were used as independent variables,and the yield of peptides after enzymolysis with gelatin was the response surface value.The optimization analysis of gelatin enzymatic process was performed.The best theoretical enzymolysis conditions were as follows:gelatin substrate concentration was 6.0%,enzyme dosage was 15.1μg/g,hydrolysis temperature was 70.00℃,pH value was 7.63 and time was 69.99 min.Fish skin gelatin peptide yield 76.54%;Finally,according to the analysis of the results and the actual combination,the optimized enzymolysis conditions for gelatin were defined as the enzymolysis temperature at 70.00℃,the gelatin concentration at 6.0%,the reaction pH at 8.00,the time at 70.0 min,and the enzyme dosage at 20.0μg/g.The actual peptide yield of enzymatic hydrolysis was 77.72%,and the error from the theoretical prediction was about 1.18%.It could be seen that the response surface model established in this experiment was reliable,had practical reference value,and could be used in practical production.
